Topic:Serological Surveys

Serological surveys for horses involve the collection and analysis of blood samples to detect the presence of antibodies or antigens related to infectious agents. These surveys are instrumental in understanding the epidemiology of diseases within equine populations, helping to identify exposure to pathogens such as viruses, bacteria, and parasites. Through serological testing, researchers can assess the prevalence of diseases, track their spread, and evaluate the effectiveness of vaccination programs. Common serological tests used in equine studies include the enzyme-linked immunosorbent assay (ELISA) and the virus neutralization test. This page compiles peer-reviewed research studies and scholarly articles that explore the methodologies, findings, and implications of serological surveys in equine health management.
Analysis of serotypes and electropherotypes of equine rotaviruses isolated in the United States.
Journal of clinical microbiology    May 1, 1991   Volume 29, Issue 5 889-893 doi: 10.1128/jcm.29.5.889-893.1991
Hardy ME, Woode GN, Xu ZC, Williams JD, Conner ME, Dwyer RM, Powell DG.Equine group A rotaviruses isolated over a 10-year period in New York State, New Jersey, Kentucky, and Texas were compared serotypically and electropherotypically. All isolates were determined to be serotype 3 by reaction with hyperimmune antiserum to the serotype 3 H-2 strain of equine rotavirus. All displayed RNA electrophoretic migration patterns related to that of the H-2 strain but distinct from that of serotype 5 strain H-1. A serologic survey of 184 mares in Kentucky, which was done to determine the incidence of H-1 and H-2 infections, showed geometric mean serum neutralizing titers to ...
[Serological studies of the recent infections of Austrian horses with the equine arteritis virus].
DTW. Deutsche tierarztliche Wochenschrift    February 1, 1991   Volume 98, Issue 2 43-45
Kölbl S, Schuller W, Pabst J.944 serum samples of horses, collected in 1988 and 1989, were examined for the occurrence of antibodies against equine arteritis virus by a microneutralizations test. In 10.9% of all sera reactors could be found. The distribution of seropositive horses varied from 4.6% (Salzburg) to 15.7% (Lower Austria). From Tyrol and Vorarlberg no samples could be obtained. It was not possible, to correlate clinical symptoms (infertility, respiratory symptoms, fever and edema) with the infection. It is assumed, that the disease appears in Austria only in a clinical inapparent form.
The occurrence of equine arteritis virus in Australia.
Australian veterinary journal    December 1, 1990   Volume 67, Issue 12 432-435 doi: 10.1111/j.1751-0813.1990.tb03052.x
Huntington PJ, Forman AJ, Ellis PM.This paper reports the first isolation of equine arteritis virus (EAV) in Australia and serological evidence of exposure to EAV in Australian horses. Twelve Standardbred stallions imported from North America were found to shed EAV in semen. One hundred and seven stallions were tested for serum antibodies to EAV and 73% of Standardbred stallions tested were seropositive as compared to 8% of Thoroughbred stallions. Serum antibody was detected in 71% of Standardbred mares, 6% of Standardbred racehorses and 1% of Thoroughbred mares and racehorses. Examination of stored serums demonstrated that EAV...
Serosurvey of horses with evidence of equine monocytic ehrlichiosis.
Journal of the American Veterinary Medical Association    November 15, 1990   Volume 197, Issue 10 1327-1332
Rikihisa Y, Reed SM, Sams RA, Gordon JC, Pretzman CI.In August 1986, an extensive serosurvey for prevalence of IgG and IgM antibodies against Ehrlichia risticii, the causative agent of equine monocytic ehrlichiosis (EME), was performed at 2 Ohio racetracks, River Downs (RD) and Beulah Park (BP). Of 840 horses at RD and 574 at BP, 13 and 20%, respectively, were IgG antibody-positive (by indirect fluorescent antibody test results), with antibody titer ranging from 1:20 to 1:10,240. The titer observed at highest frequency at both racetracks was 1:80. A higher proportion of horses was ill at RD (operating during the summer months) than at BP (winter...
[Serologic studies of the occurrence of Borrelia burgdorferi in domestic animals in Berlin (West)].
Berliner und Munchener tierarztliche Wochenschrift    November 1, 1990   Volume 103, Issue 11 374-378
Käsbohrer A, Schönberg A.The prevalence of B. burgdorferi, the causative agent of Lyme Borreliosis in humans, was determined in domestic animals living in Berlin. 189 dogs, 29 cats, 224 horses and 194 cows were investigated. Using the indirect immunofluorescence test (IFT) 5.8% of the dogs and 24.5% of the cows investigated showed a positive reaction at titres of 1:128 or higher. Horses and cats gave negative results. ELISA was more sensitive than IFT. 10.1% of the dogs, 16.1% of the horses and 66% of the local cows showed positive reaction. Domestic animals seem to be in contact with B. burgdorferi and can be a reser...
Development of an avidin-biotin dot enzyme-linked immunosorbent assay and its comparison with other serological tests for diagnosis of glanders in equines.
Veterinary microbiology    October 1, 1990   Volume 25, Issue 1 77-85 doi: 10.1016/0378-1135(90)90095-d
Verma RD, Sharma JK, Venkateswaran KS, Batra HV.A dot enzyme-linked immunosorbent assay (dot ELISA) was developed for diagnosis of glanders in equines. The test was based on the detection of IgG antibodies to Pseudomonas mallei antigens bound to nitrocellulose coated on plastic strips (dipsticks), the reaction being amplified by an avidin-biotin system with biotinylated anti-horse IgG and horseradish peroxidase-avidin D. Sera from 810 normal, six naturally infected and 48 sensitized equines were tested by this assay, and results were compared with complement fixation, indirect haemagglutination and counter-immunoelectrophoresis tests. Dot E...

Widener Hospital for Large Animals in the months of June and October, respectively, had antibody to Borrelia burgdorferi as determined by ELISA. Horses from the states of New York, Maryland, Delaware, New Jersey, and Pennsylvania were seropositive for B burgdorferi. The frequency of antibody response in horses from New Jersey was greater (P less than 0.05) than the frequency of antibody response in horses from Pennsylvania or that of horses from the other states combined. Statistically significant difference w...
[Concomitant activity of 2 bunyaviruses in horses in Argentina].
Revista Argentina de microbiologia    April 1, 1990   Volume 22, Issue 2 98-101
Cámara A, Contigiani MS, Medeot SI.A serologic survey of horses for Kairi (KRI) and Cache Valley (CV), two related Bunyaviruses, was conducted simultaneously in Cordoba and Santa Fe provinces, Argentina, during late 1983 and 1984. The prevalence of neutralizing antibodies only for KRI was 13.3% and only for CV was 40.0%; but if the total positive sera for KRI and CV were taken into account, the prevalence reached 48.3 and 75.0%, respectively. The prevalence for CV was higher than for KRI in Cordoba (p less than 0.01), but both were similar in Santa Fe province. The demonstration of seroconversion in horses of the two zones for ...
Distribution of neutralizing antibodies to California and Bunyamwera serogroup viruses in horses and rodents in California.
The American journal of tropical medicine and hygiene    March 1, 1990   Volume 42, Issue 3 282-290 doi: 10.4269/ajtmh.1990.42.282
Campbell GL, Reeves WC, Hardy JL, Eldridge BF.Neutralization tests were done on sera from 141 horses from high elevation regions of California. Antibody prevalences to Jamestown Canyon, snowshoe hare, and California encephalitis viruses in the California serogroup and Northway virus in the Bunyamwera serogroup were 55%, 43%, 18%, and 46%, respectively. In 51 horses from rural low elevation regions, seroprevalences were 31%, 35%, 35%, and 37%, respectively. Twenty-four horses from a suburban lowland area were seronegative, except for a single horse with a low titer to snowshoe hare virus. Seroprevalence to Jamestown Canyon and snowshoe har...
[New types of virus infections of domestic animals in the German Democratic Republic. 1. Serologic survey studies of the distribution of equine torovirus infections in the GDR].
Archiv fur experimentelle Veterinarmedizin    January 1, 1990   Volume 44, Issue 2 251-253
Liebermann H.Sera collected from 124 horses were checked by means of the serum neutralisation test against equine Bern virus. Torovirusspecific antibodies were recordable from 35 percent of all horses tested. These results are likely to suggest that toroviruses are widespread in the GDR and occur not only in horses but in other domestic animals and in man, as well.
Laboratory diagnosis of African horse sickness: comparison of serological techniques and evaluation of storage methods of samples for virus isolation. House C, Mikiciuk PE, Berninger ML.Five serological methods of diagnosing African horse sickness were evaluated, using a battery of serum samples from experimental horses vaccinated and challenged with each serotype of African horse sickness virus (AHSV1 through AHSV9): agar gel immunodiffusion (AGID), indirect fluorescent antibody (IFA), complement fixation (CF), virus neutralization (VN), and enzyme-linked immunosorbent assay (ELISA). The 5 tests were also compared using a panel of field samples, convalescent equine sera with antibodies to domestic equine viral diseases, and sera from horses awaiting export. The ELISA describ...

[Detection of streptococci from various serological groups in animals].
Veterinarni medicina    December 1, 1989   Volume 34, Issue 12 743-749
Havelka B, Skarková A.During the period from 1985 to 1988 we determined 228 strains of streptococci isolated from samples of various sorts of biomaterials, mainly animals. From this set of streptococci we classified 207 strains into serological groups according to the Lancefield classification and about 30% strains into species by means of serological and biochemical methods. Most of the strains were allocated to group C (37.28%) and group Q (17.39%). 89 streptococci strains originated from pigs, 40 strains from horses, 13 from cattle, 12 from dogs, 9 from poultry and 8 from coypu. The other streptococci strains or...
Monthly prevalence (in 1986) of antibody titers against equine monocytic ehrlichiosis in apparently healthy horses in Illinois.
American journal of veterinary research    November 1, 1989   Volume 50, Issue 11 1936-1939
Goetz TE, Holland CJ, Dawson JE, Ristic M, Skibbe K, Keegan KG, Johnson PJ, Schaeffer DJ, Baker GJ.The seroprevalence and seasonal trend of antibody titers against equine monocytic ehrlichiosis (Potomac horse fever) were determined in apparently healthy horses in selected areas of Illinois in 1986. Sera from 1,367 horses (6 months to 29 years old) were evaluated for the presence of antibodies against Ehrlichia risticii with indirect immunofluorescence. The majority (88%) of the horses were Thoroughbred or Standardbred racehorses. The number of horses with antibodies against E risticii was 229/1,367 (16.75%). The titers in these horses ranged from 1:10 to 1:640. As the year progressed, the n...
Antibodies to staphylococcal DNases in sera from different animal species, including humans.
Journal of clinical microbiology    November 1, 1989   Volume 27, Issue 11 2444-2447 doi: 10.1128/jcm.27.11.2444-2447.1989
Høie S, Fossum K.An agar diffusion method using microtiter plates was used to detect antibodies to the DNases produced by Staphylococcus aureus, S. intermedius, and S. hyicus. Antibodies to DNase from S. aureus were demonstrated in most of the sera from the species investigated, except dogs, only 11% of whose sera were positive. Positive titers to S. intermedius DNase were found in 84% of deg sera, 61% of Icelandic pony sera, 41% of pig sera, 21% of human sera, and 20% of cow sera but in only 2 and 4% of goat and sheep sera, respectively. Although antibodies to DNase from S. hyicus were not found in sera from ...
Epidemiology of Potomac horse fever: an investigation into the possible role of non-equine mammals.
The Veterinary record    July 22, 1989   Volume 125, Issue 4 83-86 doi: 10.1136/vr.125.4.83
Perry BD, Schmidtmann ET, Rice RM, Hansen JW, Fletcher M, Turner EC, Robl MG, Hahn NE.A serological study of antibodies to Ehrlichia risticii was carried out on 10 species of wild and domestic mammals found on or near 21 horse farms in an area of the USA in which Potomac horse fever is endemic. No antibodies were found in 133 peridomestic rodents (Norway rats and house mice), nor in 108 wild rodents (white-footed mice and meadow voles) captured on farms. Three of the six domestic animal species examined, cats, pigs and a goat, showed serological evidence of exposure to E risticii. Seropositive animals were detected on three of the 21 premises. The eight seropositive cats (of 48...
The effects of vaccination with tissue culture-derived viral vaccines on detection of antibodies to equine arteritis virus by enzyme-linked immunosorbent assay (ELISA).
Veterinary microbiology    June 1, 1989   Volume 20, Issue 2 181-189 doi: 10.1016/0378-1135(89)90041-2
Cook RF, Gann SJ, Mumford JA.An enzyme-linked immunosorbent assay (ELISA) was developed for the detection of serum antibodies to equine arteritis virus (EAV). Results from this assay produced a good correlation with results from virus neutralisation tests in horses which had not been regularly vaccinated with commercially available mammalian tissue culture-derived viral vaccines. Vaccination of some horses with tissue culture-derived vaccines induced the formation of antibodies to bovine serum. These antibodies reacted with the bovine protein contaminants in the EAV ELISA antigen, producing false-positive results. Non-vir...
Complement fixing antibodies against arboviruses in horses at Lagos, Nigeria.
Revue d'elevage et de medecine veterinaire des pays tropicaux    January 1, 1989   Volume 42, Issue 3 321-325
Olaleye OD, Oladosu LA, Omilabu SA, Baba SS, Fagbami AH.Sixty-two sera horse collected from two stables at Lagos, Nigeria, were tested for complement fixing antibody to 8 arbovirus antigens; Chikungunya, Igbo-Ora, Yellow fever, Wesselsbron, West Nile, Potiskum, Uganda S and Rift Valley fever. Ten per cent of the horse sera examined contained CF antibody to one or more of the test antigens and indicated considerable arbovirus activity in the two stables. Reactions with flavivirus antigens were most common and the highest antibody titres were obtained with Wesselsbron and Yellow fever viruses. Eleven per cent of the sera tested reacted with alphaviru...

Godsey MS, Amoo F, Yuill TM, Defoliart GR.A serologic survey and experimental virus transmission studies were done to assess the role of domestic animals as amplifier hosts of La Crosse (LACV) and Jamestown Canyon (JCV) viruses. Serum from 319 cows, 88 dogs, 122 equines, 47 swine, 10 goats, and 4 cats were tested for neutralizing antibody to LACV, JCV, trivittatus (TVTV), and snowshoe hare (SSHV) viruses. Antibody prevalences of LACV, TVTV, and SSHV were less than 10% in all species. Antibody to JCV was detected in all species except cats. Prevalence ranged from 10% in goats and swine to 29% in dogs. No age-associated trends in JCV pr...
Pathogenicity for horses of original Sagiyama virus, a member of the Getah virus group.
Veterinary microbiology    August 1, 1988   Volume 17, Issue 4 367-373 doi: 10.1016/0378-1135(88)90050-8
Kumanomido T, Kamada M, Wada R, Kenemaru T, Sugiura T, Akiyama Y.Sagiyama virus is a member of the Getah virus group. Its pathogenicity for horses was examined. All the horses infected with the original 4 strains of Sagiyama virus (M6/Mag 33, Mag 121, Mag 132 and Mag 258) developed pyrexia ranging from 39.0 to 40.0 degrees C. Other clinical signs, characterized by eruptions, edema in the hind legs, enlargement of the submandibular lymph node and mild leukopenia, were also manifested. Viremia occurred 1-4 days post-inoculation (p.i.). Virus was recovered from spleen, liver, lung and various lymph nodes of a horse autopsied on Day 4 p.i. The maximum titer of ...
An investigation into alternative methods for the serodiagnosis of dourine.
The Onderstepoort journal of veterinary research    June 1, 1988   Volume 55, Issue 2 117-119
Williamson CC, Stoltsz WH, Mattheus A, Schiele GJ.The complement fixation test (CFT), indirect fluorescent antibody test (IFAT), card agglutination test for trypanosomiasis (CATT) and enzyme-linked immunosorbent assay (ELISA) were compared in their application to the serological diagnosis of Trypanosoma equiperdum infection in 43 horses. The CFT remains a reliable test for dourine, especially in countries where other members of the subgenus Trypanozoon do not occur. The IFAT is a good 'back-up' test, but, requiring skilled operators it has the disadvantage of making it labour intensive, and interpretation of results subjective. This makes it ...

Epidemiologic studies of Lyme disease in horses and their public health significance.
Annals of the New York Academy of Sciences    January 1, 1988   Volume 539 244-257 doi: 10.1111/j.1749-6632.1988.tb31858.x
Cohen D, Bosler EM, Bernard W, Meirs D, Eisner R, Schulze TL.A serologic survey of horses in the New Jersey-Pennsylvania area demonstrated that about 10% (6.2-14.2%) have significant levels of serum antibody to Borrelia burgdorferi. However, in a highly endemic area of central New Jersey, up to 60% of the mares and yearlings samples on one farm were seropositive. In 1983, sera from this same farm exhibited only 12% positives in mares and 35% positives in yearlings. Longitudinal studies of paired sera obtained from individual yearlings over a 6-month period in 1985 showed that 34% of them declined during the period. A new clinical syndrome associated wit...
Sero-epizootiological study of racehorses with pyrexia in the training centers of the Japan Racing Association.
Nihon juigaku zasshi. The Japanese journal of veterinary science    December 1, 1987   Volume 49, Issue 6 1087-1096 doi: 10.1292/jvms1939.49.1087
Sugiura T, Matsumura T, Fukunaga Y, Hirasawa K.A sero-epizootiological study was conducted on horses which showed clinical pyrexia at two training centers, each of which maintained 1, 200 to 2, 100 racehorses for training, of the Japan Racing Association. It continued from 1980 to 1985 to clarify the cause of pyrexia, so that measures might be considered for the prevention of infectious disease. A total of 3, 849 horses were found to be affected with pyrexia for the 6 years. Of them, 2, 852 horses were tested to equine herpesvirus type 1 (EHV-1), equine rhinovirus type 1 (ERhV-1), rotavirus and equine adenovirus (EAdV) by collecting paired...
Diagnosis of Rhodococcus equi infection in foals by the agar gel diffusion test with protein antigen.
Veterinary microbiology    October 1, 1987   Volume 15, Issue 1-2 105-113 doi: 10.1016/0378-1135(87)90135-0
Nakazawa M, Isayama Y, Kashiwazaki M, Yasui T.A protein antigen that reacted in the agar gel diffusion (AGD) test and which had equi factor(s) activity, was partially purified from the culture supernatant of Rhodococcus equi by successive column chromatography on diethylaminoethyl cellulose and Sepharose 4B. Employing a standard foal serum, the concentration of this antigen was adjusted for the AGD test. Optimal dilutions of the antigen reacted in the AGD test with sera from foals naturally infected with serologically different R. equi. The antigen prepared was considered suitable for use in field surveys of R. equi infection. Accordingly...
Leptospirosis in horses in Ontario.
Canadian journal of veterinary research = Revue canadienne de recherche veterinaire    October 1, 1987   Volume 51, Issue 4 448-451
Kitson-Piggot AW, Prescott JF.Sera from Thoroughbred and Standardbred horses in southwest Ontario were tested for antibody to seven Leptospira interrogans serovars (autumnalis, bratislava, canicola, grippotyphosa, hardjo, icterohaemorrhagiae, pomona), using the microscopic agglutination test. There was significantly higher seroprevalence of bratislava than of other serovars, in which prevalence was low. Seroprevalence of bratislava increased significantly with age; only 5% of two to three year old horses had titers greater than or equal to 1:80 compared to 52% of horses older than seven years. Eight of 16 foals from two fa...
