Topic:Stallion

Stallions are adult male horses that have not been gelded and are often used for breeding purposes. They exhibit distinct behavioral and physiological characteristics compared to mares and geldings. Stallions are known for their strong musculature, prominent neck, and often more pronounced mane and tail. Their behavior can be more aggressive and territorial, especially during the breeding season, as they naturally compete for mating opportunities. This topic explores the anatomy, behavior, and management practices specific to stallions, as well as their role in equine reproduction. The page provides access to peer-reviewed research studies and scholarly articles that investigate various aspects of stallion health, fertility, and behavior, contributing to the understanding and management of these horses in equine science.
An Investigation of Equine Sperm Quality Following Cryopreservation at Low Sperm Concentration and Repeated Freeze-Thawing.
Journal of equine veterinary science    December 1, 2022   Volume 120 104167 doi: 10.1016/j.jevs.2022.104167
Morse-Wolfe B, Bleach E, Kershaw C.Stallion spermatozoa are typically cryopreserved at 200 to 300 million sperm/ml; however recent advances such as intracytoplasmic sperm injection (ICSI) requires only one spermatozoon, wasting many, after thawing a whole straw. Cryopreserving at concentrations less than the current standard or refreezing thawed spermatozoa could maximize the use of genetically valuable animals and reduce waste. This investigation aimed to identify if lowering the sperm concentration for cryopreservation affected post-thaw quality after one and two freeze-thaw cycles. Nine ejaculates were collected from three f...
Effects of egg yolk level, penetrating cryoprotectant, and pre-freeze cooling rate, on the post-thaw quality of stallion sperm.
Animal reproduction science    December 1, 2022   Volume 248 107162 doi: 10.1016/j.anireprosci.2022.107162
Hernández-Avilés C, Ramírez-Agámez L, Varner DD, Love CC.The current study determined the effect of the egg-yolk (phospholipid source) level (egg yolk [20% EY] vs. skim-milk + egg yolk [SM + 2% EY]), cryoprotectant (glycerol [Gly] vs. glycerol + methylformamide [Gly + MF]), and pre-freeze cooling rate (-0.1 vs. -1 vs. -5 °C/min) on post-thaw stallion sperm quality. In Experiment 1, ejaculates (n = 27) from 9 stallions (3 ejaculates each) with varied sperm quality (High, Average, or Low) were frozen in EY-Gly, SMEY-Gly, EY-Gly + MF, or SMEY-Gly + MF extenders. Sperm in each group were cooled from 22° to 5°C using either -0.1 °C/min or -1 Â...

Animal reproduction    November 28, 2022   Volume 19, Issue 4 e20220010 doi: 10.1590/1984-3143-AR2022-0010
Aguiar CS, Barros CHSC, Machado WM, Allaman IB, Leite AO, Barbosa LP, Snoeck PPDN.The aim of this study was to evaluate the association of different concentrations of Trolox and the addition of a fixed concentration of DHA in the freezing of semen of Mangalarga Marchador stallions. To that end, 16 ejaculates were frozen in the following extenders: E1) BotuCrio (BC; Control); E2) BC + 50 ngml DHA + 30 µM Trolox (BCDHA30T); E3) BC + 50 ngml DHA + 40 µM Trolox (BCDHA40T); E4) BC + 50 ngml DHA + 50 µM Trolox (BCDHA50T). All the tested extenders were similar in preserving different kinematic parameters, cell functional integrity, compacted DNA, and high and intermediate mitoc...
Penicillin and amikacin mixture has bactericidal activity equivalent to gentamicin, tylosin, lincomycin and spectinomycin mixture in equine frozen semen.
Reproduction in domestic animals = Zuchthygiene    November 25, 2022   Volume 58, Issue 2 349-357 doi: 10.1111/rda.14294
Brito LFC, Loomis PR, Klohonatz KM, Althouse GC.Neat stallion semen can contain a variety of microorganisms, some of which may impair sperm quality and/or cause infection of the mares' reproductive tract. For this reason, antibiotics are commonly added to semen extenders. A combination of gentamicin, tylosin, lincomycin and spectinomycin (GTLS) has been recommended for use, but there are no reports on the use of this mixture in equine semen extender. Penicillin and amikacin (PA) are safe for preserving sperm quality while effectively controlling bacterial growth in equine cooled stored semen, but data on frozen semen are scarce. Therefore, ...
Action of swim-up and caffeine on equine frozen sperm.
Animal reproduction    November 21, 2022   Volume 19, Issue 4 e20220056 doi: 10.1590/1984-3143-AR2022-0056
Alves NC, Diniz SA, Viegas RN, Arigoni AL, Freitas MM, Lana ÂQ, Lagares MA.Cryopreservation of equine semen is crucial to semen commercialization. However, it reduces sperm motility and longevity. Thus, sperm selection methods and addition of motility-activating substances to sperm, such as caffeine, may improve sperm quality of equine frozen semen. The objective of the current work was to evaluate the effects of caffeine on recovery and quality parameters of frozen-thawed sperm subjected to swim-up selection to be used in intracytoplasmic sperm injection (ICSI) in assisted reproductive techniques. Stallion semen were frozen and after thawing different caffeine conce...
Determining Objective Parameters to Assess Gait Quality in Franches-Montagnes Horses for Ground Coverage and Over-Tracking – Part 2: At Trot.
Journal of equine veterinary science    November 20, 2022   Volume 120 104166 doi: 10.1016/j.jevs.2022.104166
Gmel AI, Haraldsdóttir EH, Bragança FMS, Cruz AM, Neuditschko M, Weishaupt MA.In gait quality assessments of horses, stride length (SL) is visually associated with spectacular movements of the front limbs, and described as ground coverage, while the movement of the hind limb under the body is supposedly essential to a longer over-tracking distance (OTD). To identify movement patterns with strong associations to SL and OTD, limb and body kinematics of 24 Franches-Montagnes (FM) stallions were measured with 3D optical motion capture (OMC) on a treadmill during an incremental speed test at trot (3.3-6.5 m/s). These measurements were correlated to the scores of ground cover...
OXIDATIVE STRESS AND REPRODUCTIVE FUNCTION: Oxidative stress and the long-term storage of horse spermatozoa.
Reproduction (Cambridge, England)    November 18, 2022   Volume 164, Issue 6 F135-F144 doi: 10.1530/REP-22-0264
Peña FJ, Gibb Z.The growing understanding of the mechanisms regulating redox homeostasis in the stallion spermatozoa, together with its interactions with energetic metabolism, is providing new clues applicable to the improvement of sperm conservation in horses. Based on this knowledge, new extenders, adapted to the biology of the stallion spermatozoa, are expected to be developed in the near future. The preservation of semen either by refrigeration or cryopreservation is a principal component of most animal breeding industries. Although this procedure has been successful in many species, in others, substantia...
Would the Cephalic Development in the Purebred Arabian Horse and Its Crosses Indicate a Paedomorphic Process?
Animals : an open access journal from MDPI    November 16, 2022   Volume 12, Issue 22 3168 doi: 10.3390/ani12223168
Salamanca-Carreño A, Parés-Casanova PM, Monroy-Ochoa NI, Vélez-Terranova M.This study examined paedomorphosis in PAH and F1 crossbreds. A sample of 99 horses was selected from 40 different breeders and consisted of three groups: stallions ( = 16), mares ( = 53), and geldings ( = 30), ranging from 10 months to 27 years in age. All horses presented a concave celloid lateral left head profile in the acquired photographic images. The hypothesis proposed in this study suggested the lateral profile of the head in juveniles was representational in the adult form due to the neonate's facial bones (part of the splanchnocranium) developing at a different rate to those of the s...
The stallion sperm acrosome: Considerations from a research and clinical perspective.
Theriogenology    November 14, 2022   Volume 196 121-149 doi: 10.1016/j.theriogenology.2022.11.012
Hernández-Avilés C, Ramírez-Agámez L, Varner DD, Love CC.During the fertilization process, the interaction between the sperm and the oocyte is mediated by a process known as acrosomal exocytosis (AE). Although the role of the sperm acrosome on fertilization has been studied extensively over the last 70 years, little is known about the molecular mechanisms that govern acrosomal function, particularly in species other than mice or humans. Even though subfertility due to acrosomal dysfunction is less common in large animals than in humans, the evaluation of sperm acrosomal function should be considered not only as a complementary but a routine test whe...
Kallikrein proteoforms and reproductive parameters in stallion are conditioned by climate.
Scientific reports    November 4, 2022   Volume 12, Issue 1 18690 doi: 10.1038/s41598-022-21350-w
Senra RL, Ramírez-López CJ, Magalhães-Júnior MJ, Neves JGDS, Barros E, Waddington B, Guimarães SEF, Guimarães JD, Baracat-Pereira MC.Horses are seasonal polyoestrous animals, and the photoperiod is the main factor modulating their reproductive activity. There is no consensus on the andrological and biochemical factors that influence breeding seasonality. To assess the involvement of climate in reproduction, Mangalarga Marchador stallions were monitored over 1 year regarding semen quality and seminal plasma proteome. Here, we show that kallikrein (KLKs) proteoforms in seminal plasma are involved in climate conditioning of reproduction. During the breeding season, greater abundance and different types of KLKs occurred simult...
Changes in Management, Welfare, Emotional State, and Human-Related Docility in Stallions.
Animals : an open access journal from MDPI    October 30, 2022   Volume 12, Issue 21 2981 doi: 10.3390/ani12212981
Popescu S, Lazar EA, Borda C, Blaga Petrean A, Mitrănescu E.Despite an increase in awareness of their essential needs, many stallions continue to be kept in conditions limiting their social interactions and movement. To supplement the studies which highlight the effects of these practices on selected aspects of equine mental and physical wellbeing, we aimed to monitor a group of 32 adult intact stallions during their transition from tethered housing with limited outdoor access to free group housing through the lens of their overall welfare, perceived emotional status, and docility toward humans. Over three visits (before the management change, two week...
Sperm factors associated with the production of equine blastocysts by intracytoplasmic sperm injection (ICSI) using frozen/thawed semen.
Theriogenology    October 17, 2022   Volume 195 85-92 doi: 10.1016/j.theriogenology.2022.10.014
Ramírez-Agámez L, Hernández-Avilés C, Varner DD, Love CC.Intracytoplasmic Sperm Injection (ICSI) using frozen/thawed sperm is a common procedure to obtain embryos from fertile or subfertile mares and stallions. Stallion-associated factors that impact the efficiency of ICSI have been studied less than those associated with the mare. Three experiments were conducted: Experiment 1: the effect of freezing extender composition and cryoprotectant; Experiment 2: the effect of sperm exposure to seminal plasma prior to freezing (ejaculated vs. epididymal sperm; two-freeze/thaw cycles each); and Experiment 3: the effect of sperm morphologic feature used for f...

Heliyon    October 17, 2022   Volume 8, Issue 10 e11128 doi: 10.1016/j.heliyon.2022.e11128
Waheed MM, Meligy A, Alhaider AK, Ghoneim IM.Seminal plasma contains several microelements like Zn, Fe, Se, and Cu that affect sperm motility and male fertility. Biochemical evaluation of seminal plasma trace elements is important for assessing fertility and diagnosing male infertility. Unassigned: The present study was designed to evaluate the effect of seminal fluid trace elements on sperm parameters and fertility in Arabian horses. Unassigned: Ninety-four ejaculates from 25 Arabian stallions (4-27 years old) were used to investigate the effect of seminal fluid trace elements on semen parameters and fertility. Data divided according to...
Copy number variation of horse Y chromosome genes in normal equine populations and in horses with abnormal sex development and subfertility: relationship of copy number variations with Y haplogroups.
G3 (Bethesda, Md.)    October 14, 2022   Volume 12, Issue 12 jkac278 doi: 10.1093/g3journal/jkac278
Castaneda C, Radović L, Felkel S, Juras R, Davis BW, Cothran EG, Wallner B, Raudsepp T.Structural rearrangements like copy number variations in the male-specific Y chromosome have been associated with male fertility phenotypes in human and mouse but have been sparsely studied in other mammalian species. Here, we designed digital droplet PCR assays for 7 horse male-specific Y chromosome multicopy genes and SRY and evaluated their absolute copy numbers in 209 normal male horses of 22 breeds, 73 XY horses with disorders of sex development and/or infertility, 5 Przewalski's horses and 2 kulans. This established baseline copy number for these genes in horses. The TSPY gene showed the...
Screening for Taylorella equigenitalis in Equine Semen: An Exploratory Study.
Journal of equine veterinary science    October 13, 2022   Volume 119 104138 doi: 10.1016/j.jevs.2022.104138
Mawhinney I, Davis N, Carson T, Torrens N, Wales A.The study examined and compared the sensitivity of culture and a quantitative PCR assay for screening equine semen for the presence of Taylorella equigenitalis (CEMO). Chilled semen samples, both raw and treated with extender, from two stallions were spiked with the organism at seven or 23 days postejaculation and prepared in serial dilutions. Culture of the 7-day raw semen readily detected CEMO at all dilutions, but extended semen yielded counts that were two log cycles lower at equivalent dilutions, with the organism being nearly undetectable at the maximal dilutions. By contrast, PCR sensit...
Liquid storage of stallion spermatozoa – Past, present and future.
Animal reproduction science    October 6, 2022   Volume 247 107088 doi: 10.1016/j.anireprosci.2022.107088
Clulow J, Gibb Z.The ability to store stallion spermatozoa between the events of semen collection and insemination has facilitated improved rates of gain in selective breeding programs by enabling the transport of spermatozoa. While cryopreservation is the only viable means of storing spermatozoa for long intervals, the higher costs and reduced fertility of cryopreserved spermatozoa have led to most breeders opting to use liquid stored spermatozoa. Stallion spermatozoa is commonly cooled during liquid storage (approximately 4-5 °C), and there has been an enormous body of research dedicated to development of ...
Evidence of maternal and paternal age effects on speed in thoroughbred racehorses.
Royal Society open science    October 5, 2022   Volume 9, Issue 10 220691 doi: 10.1098/rsos.220691
Sharman P, Young AJ, Wilson AJ.Effects of parental age on offspring viability have been reported in a wide range of species. However, to what extent parental age influences offspring traits beyond viability remains unclear. Moreover, previous research has primarily focused on maternal age effects. The purpose of this study was to test for paternal and maternal age effects on offspring speed in thoroughbred racehorses. We analysed over 900 000 race performances by over 100 000 horses on British racecourses between 1996 and 2019. With knowledge of the age of all 41 107 dams and 2 887 sires at offspring conception, we jointly ...
Exploring the metabolome of seminal plasma in two different horse types: Light versus draft stallions.
Reproduction in domestic animals = Zuchthygiene    October 3, 2022   Volume 58, Issue 1 109-116 doi: 10.1111/rda.14270
Bazzano M, Zhu C, Laus F, Giambattista AD, Laghi L.The application of the 'omics' studies in the field of animal reproduction has been aimed at identifying novel biomarkers of fertility since the last few years. When assessing reproductive efficiency in horses, breed should also be taken into account as it can influence semen quality and fertility. Considering the growing interest in metabolomic analysis to evaluate male fertility, we aimed to investigate the metabolomic profile of seminal plasma in two different horse breeds. Twelve healthy stallions, n.6 American Quarter Horse (AQH) and n.6 Italian Draft Horse (IDH) stallions, regularly used...

Journal of equine veterinary science    September 29, 2022   Volume 118 104132 doi: 10.1016/j.jevs.2022.104132
van Heule M, Verstraete M, Blockx Z, De Blende P, Dini P, Daels P.Cooled storage of semen after thawing can expand the use of frozen semen, providing the possibility of thawing and evaluating the semen at the storage site and subsequently shipping the semen. Our objectives were (1) to examine the motility and viability of frozen-thawed semen after cooled storage and (2) to compare two cooled-storage protocols for frozen-thawed semen. The samples (n = 31) were either placed immediately in a passive cooling box for 8 or 24 hours (CB) or placed in a refrigerator at 4°C for 30 minutes and then transferred to a passive cooling box (REF). Total and progressive ...

Theriogenology    September 24, 2022   Volume 194 27-34 doi: 10.1016/j.theriogenology.2022.09.011
Gautier C, Aurich J, Kaps M, Okada CTC, Wagner LH, Melchert M, Aurich C.In stallions temporarily not intended for breeding, reversible suppression of testicular function by vaccination against GnRH can be of interest. In the present study, effects of GnRH agonist treatment on the resumption of testicular function after GnRH vaccination were investigated. Testis size, testosterone release, semen characteristics and behavior were evaluated. We hypothesized that GnRH agonist treatment would restore testicular function. Shetland stallions were assigned to an experimental and a control group (n = 6 each). Experimental stallions were GnRH-immunized twice, four weeks a...

Animal reproduction science    September 23, 2022   Volume 247 107076 doi: 10.1016/j.anireprosci.2022.107076
Quirino M, Jakop U, Mellagi APG, Bortolozzo FP, Jung M, Schulze M.For a more practically applicable analysis of different sperm characteristics, this study aimed to develop a 5-color flow cytometry (FC) panel to concurrently analyze four sperm parameters in liquid boar and stallion semen, using also a DNA-marker for selecting sperm cell events. From each of thirty extended boar semen doses and twelve stallion semen doses, six aliquots were taken. For evaluating mitochondrial activity (A), degree of lipid disorder of plasma membrane (B), integrity of plasma membrane (C), acrosomal status (D) and marking DNA (E), five aliquots were individually stained with Rh...
Bacterial killing activity and lysozymes: A stable defence mechanism in stallion seminal plasma?
Reproduction in domestic animals = Zuchthygiene    September 21, 2022   Volume 58, Issue 1 73-80 doi: 10.1111/rda.14260
Jakop U, Hensel B, Czirják GÁ, Quirino M, Schröter F, Jung M, Schulze M.During insemination, bacterial contamination of the ejaculate can lead to reduced sperm quality and transmission of pathogens to the female, thus should be avoided. The semen of a variety of animal taxa possess antimicrobial properties against a wide range of bacterial species through antimicrobial molecules, such as lysozyme, but their variance and the factors influencing it are unknown for most species. In this study, the antibacterial defence (bacterial killing activity (BKA) against Escherichia (E.) coli and Staphylococcus (S.) aureus as well as lysozyme concentration) was studied in semin...
Dynamics of acylcarnitines, hypoglycin A, méthylènecyclopropylglycine and their metabolites in a Kladruber stallion with atypical myopathy.
The veterinary quarterly    September 18, 2022   Volume 42, Issue 1 183-191 doi: 10.1080/01652176.2022.2126537
Jahn P, Dobešová D, Brumarová R, Tóthová K, Kopecká A, Friedecký D.Equine atypical myopathy (AM also referred to as multiple acyl-CoA dehydrogenases deficiency [MADD]) is thought to be caused by toxins metabolized from hypoglycin A (HGA) and méthylènecyclopropylglycine (MCPrG). HGA is contained in the seeds and seedlings of the sycamore tree (Acer pseudoplatanus); MCPrG has so far only been confirmed in seeds. Among other things, these substances can disrupt the fatty acids β-oxidation pathway with the subsequent accumulation of certain acylcarnitines. The tentative diagnosis is based on anamnesis and clinical signs and can be verified by the detection of ...
Imaging flow cytometry to characterize the relationship between abnormal sperm morphologies and reactive oxygen species in stallion sperm.
Reproduction in domestic animals = Zuchthygiene    September 14, 2022   Volume 58, Issue 1 10-19 doi: 10.1111/rda.14241
Bulkeley E, Santistevan AC, Varner D, Meyers S.Low levels of intracellular reactive oxygen species (ROS) are essential for normal sperm function and are produced by sperm mitochondria as a byproduct of metabolism, but in excess, ROS can cause catastrophic cellular damage and has been correlated with infertility, poor sperm motility and abnormal morphology in humans. Stallion sperm motility is fueled predominantly by oxidative phosphorylation-produced ATP, requiring high basal rates of mitochondrial function. Consequently, whether elevated ROS production by stallion sperm is an indicator of dysfunctional or highly motile cells has been deba...
Dataset of the sperm proteome of stallions with different motility.
Data in brief    September 5, 2022   Volume 45 108578 doi: 10.1016/j.dib.2022.108578
Gaitskell-Phillips G, Martín-Cano FE, Ortiz-Rodríguez JM, Silva-Rodríguez A, da Silva-Álvarez E, Gil MC, Ortega-Ferrusola C, Peña FJ.This paper provides a detailed set of data on how the stallion sperm proteome differs among stallions with different sperm motilities, although within normal ranges. Findings distinguish proteins that may help to identify stallions of superior sperm motility. Sperm proteins were analyzed using a UHPLC/MS/MS system comprising of an Agilent 1290 infinity series UHPLC coupled to an Agilent 6550 Q-TOF mass spectrometer (Agilent Technologies, Santa Clara, CA, USA). These data can be used to disclose potential targets to identify good sperm samples and to study specific pathways involved in the regu...
Seasonal changes in the expression of molecular markers of stallion germ cells.
Journal of equine veterinary science    August 24, 2022   Volume 118 104109 doi: 10.1016/j.jevs.2022.104109
Shakeel M, Jung H, Yoon D, Yoon M.The economic impacts of infertility and subfertility of stallions greatly influence the horse breeding industry. Self-renewal and differentiation of spermatogonial stem cells are the initial processes to maintain an adequate sperm population. Thus, understanding these processes may provide useful information to reveal the causes and remedies of subfertile and infertile stallions. Stallions are seasonal breeders. About 50% of the sperm population is reduced during the non-breeding season (NBS) in stallions. The seasonal regulation of spermatogenesis renders stallions as ideal models to understa...
Characterization of sperm cell membrane charge and selection of high-quality sperm using microfluidics in stallions.
Theriogenology    August 17, 2022   Volume 192 1-8 doi: 10.1016/j.theriogenology.2022.08.014
Orsolini MF, Verstraete MH, van Heule M, Orellana D, Ortega A, Meyers S, Dini P.Intracytoplasmic sperm injection (ICSI) is the only method for in vitro embryo production (IVP) in horses. Besides oocyte developmental competence, the outcome of IVP is also highly dependent on sperm quality. Therefore, it is not only essential to employ superior methods of selecting high quality sperm, but also to be able to characterize which quantifiable properties of sperm quality are most indicative of its fertility. In men, a net negative surface charge, estimated by zeta potential (ZP) is highly correlated with sperm quality and in vitro embryo developmental outcomes. However, there ...
Differential expression of glycans in the urothelial layers of horse urinary bladder.
Annals of anatomy = Anatomischer Anzeiger : official organ of the Anatomische Gesellschaft    August 17, 2022   Volume 244 151988 doi: 10.1016/j.aanat.2022.151988
Desantis S, Santamaria N, Zizza S, Accogli G, Mastrodonato M, Scillitani G, Mentino D.Urothelium is a multilayer epithelium covering the inner surface of the urinary bladder that acts as a blood-urine barrier and is involved in maintaining the wellbeing of the whole organism. Glycans serve in the maturation and differentiation of cells and thus play a key role in the morphology and function of the multilayered epithelium. The aim of the present study was to examine the glycoprotein pattern of the horse urinary bladder urothelium by lectin histochemistry. Methods: The study involved urinary bladders from four horse stallions. Tissue sections were stained with a panel of eleven l...
Equine Gastric Squamous Cell Carcinoma in a Friesian Stallion.
Journal of equine veterinary science    July 29, 2022   Volume 117 104087 doi: 10.1016/j.jevs.2022.104087
Straticò P, Razzuoli E, Hattab J, Guerri G, Celani G, Palozzo A, Bonanni D, Fruscione F, Varasano V, Petrizzi L, Marruchella G.A 7-year-old Friesian stallion with a history of oesophageal stenosis, weight loss, inappetence, and recurrent hyperthermia was referred for gastroscopy. The stomach mucosa surrounding the oesophageal opening showed a large, necrotic, and ulcerated mass. On post-mortem examination, a very large, cauliflower-like neoplasm was seen, affecting non-glandular gastric mucosa. Nodular lesions were observed, scattered on the omentum, the spleen, and the liver. Microscopic findings allowed the diagnosis of gastric squamous cell carcinoma with abdominal metastasis. Biomolecular investigations demonstrat...
