Topic:In Vivo

In vivo studies involving horses refer to research conducted within living organisms to understand various biological processes, disease mechanisms, and therapeutic interventions in equine species. These studies provide insights into the physiological and pathological responses of horses under natural conditions. In vivo research encompasses a range of investigations, including pharmacokinetics, disease modeling, and the evaluation of treatment efficacy. It often involves the use of controlled environments to simulate real-life scenarios while observing the effects of specific variables on equine health. This page gathers peer-reviewed research articles and scholarly studies that explore the methodologies, findings, and implications of in vivo research in horses, contributing to the advancement of equine science and veterinary medicine.
Effect of time and storage temperature on cholinesterase activity in blood from normal and organophosphorus insecticide-treated horses. Plumlee KH, Richardson ER, Gardner IA, Galey FD.Delays between time of sampling and time of testing are common; therefore, the length of time that blood can be stored at various temperatures was evaluated for effects on cholinesterase activity. Six horses were treated with 16 g of trichlorfon per os, 6 horses were treated with 15 g of dichlorvos per os, and 10 horses were untreated controls. The cholinesterase activity in whole blood from each horse was measured using an adaptation of the Ellman colorimetric method. The blood from each horse was then divided into 3 groups and stored at 5 C (refrigerated), 20 C (room temperature), or 38 C (i...
Control of cambendazole-resistant small strongyles (Population S) with oxibendazole in a pony band: an 8 year field test (1984-1992).
Veterinary parasitology    April 1, 1994   Volume 52, Issue 3-4 271-277 doi: 10.1016/0304-4017(94)90118-x
Lyons ET, Drudge JH, Tolliver SC, Swerczek TW, Stamper S, Granstrom DE.Studies in a band of ponies harboring Population S benzimidazole-resistant small strongyles were initiated in 1974 and have continued for 18 years. Treatment (bimonthly) was with cambendazole for the first 4 years and with oxibendazole (OBZ) for the next 14 years. Data on the first 10 years have been published. The present investigation includes the last 8 years (4 October 1984-11 September 1992), which are the seventh through fourteenth years, of treatment with OBZ. Pre- and posttreatment mean counts of strongyle eggs (epg) and larvae (lpg) per gram of feces were determined biweekly during th...
Ultracytochemical study of multiple dose effect of monastral blue uptake by equine pulmonary intravascular macrophages (PIMs).
Journal of submicroscopic cytology and pathology    April 1, 1994   Volume 26, Issue 2 235-243
Singh B, Minhas KJ, Atwal OS.The pulmonary intravascular macrophages (PIMs) of the ponies possess all the characteristics of a fully differentiated resident macrophage, which forms adhesive plaques with the capillary-endothelial cells. In addition, it has a unique surface coat which does not conform to the concept of a carbohydrate rich membrane bound glycocalyx generally associated with macrophages. We studied the responses of these cells especially in the context of its globular surface coat to multiple doses of MB intravenously: 0.2 ml/kg body copper tracer substance. The ponies were treated with three doses of MB intr...
In vivo replicative status and envelope heterogeneity of equine infectious anemia virus in an inapparent carrier.
Journal of virology    April 1, 1994   Volume 68, Issue 4 2777-2780 doi: 10.1128/JVI.68.4.2777-2780.1994
Kim CH, Casey JW.The distribution and replicative status of equine infectious anemia virus (EIAV) DNA in the tissues of a well-characterized inapparent carrier horse were established by using the PCR technique. The EIAV pol region could be amplified in all of the tissues tested, including the cerebellum and periventricular tissue, at concentrations approximately 10(5)-fold less than in the same tissue from an acutely infected horse. Further analysis of the EIAV genome, with primer pairs diagnostic for sequential stages of reverse transcription, suggests that EIAV DNA in the brain, liver, and lymph nodes was in...
A comparison of ovine and equine antivenoms.
Toxicon : official journal of the International Society on Toxinology    April 1, 1994   Volume 32, Issue 4 427-433 doi: 10.1016/0041-0101(94)90294-1
Sjostrom L, al-Abdulla IH, Rawat S, Smith DC, Landon J.Commercial antivenoms produced in horses were compared with monospecific antivenoms raised in sheep against Crotalus durissus terrificus, Crotalus atrox, Crotalus adamanteus, Micrurus fulvius fulvius, Naja naja, Naja kaouthia, Echis ocellatus, Vipera lebetina deserti, Vipera berus berus and Vipera ammodytes ammodytes venom. Antibodies raised by immunizing sheep with C. d. terrificus venom were more effective than their equine counterparts in preventing lethal toxicity in mice (ED50), in inhibiting the venom's pharmacological effects (haemolysis, platelet aggregation and coagulation), and in ne...
Babesia equi erythrocytic stage continuously cultured in an enriched medium.
The Journal of parasitology    April 1, 1994   Volume 80, Issue 2 232-236
Holman PJ, Chieves L, Frerichs WM, Olson D, Wagner GG.Babesia equi was continuously cultured through 90 passages in an enriched chemically defined basal medium (HL-1) supplemented with 20% fetal bovine serum and serum replacement factors, including lipid-rich bovine serum albumin, bovine insulin, and human transferrin. Cryopreservation and subsequent recovery of B. equi were easily achieved. Inoculation of a splenectomized and an intact horse with cultured infected erythrocytes resulted in parasitemias and B. equi in vitro reisolation from both animals. In vitro forms of the parasite resembled in vivo forms. After establishment, parasitemias of 1...
Comparison of the effects of ketoprofen and flunixin meglumine on the in vitro response of equine peripheral blood monocytes to bacterial endotoxin. Jackman BR, Moore JN, Barton MH, Morris DD.The purpose of this study was to investigate the in vitro effects of flunixin meglumine, a cyclo-oxygenase inhibitor, and ketoprofen, a reported cyclo-oxygenase and lipoxygenase inhibitor, on the synthesis of cyclo-oxygenase end-products thromboxane B2 and prostaglandin E2, lipoxygenase derived 12-hydroxyeicosatetraenoic acid, tumor necrosis factor and tissue factor. Six adult horses were each randomly administered flunixin meglumine (1.1 mg/kg) or ketoprofen (2.2 mg/kg) intravenously every 12 hours with the drug treatments separated by two weeks. Blood samples were obtained prior to initiatin...

Research in veterinary science    March 1, 1994   Volume 56, Issue 2 256-258 doi: 10.1016/0034-5288(94)90113-9
Baker SJ, Gerring EL.Impedance epigastrography was used to measure gastric emptying rates on two occasions in each of three foals. After smoothing of the raw data, emptying of water appeared to obey an exponential model. The mean emptying rate constant (SEM) was 0.171 +/- 0.038 min-1 (mean emptying half time 4.9 +/- 1.0 min).
An ultrastructural study of intranuclear bodies in granular epithelial cells of the equine small intestine.
Equine veterinary journal    March 1, 1994   Volume 26, Issue 2 156-158 doi: 10.1111/j.2042-3306.1994.tb04357.x
Kaup FJ, Deegen E.Gastrointestinal disorders and colic are common problems of horses. In spite of its clinical importance, only a few reports deal with the normal morphology of the equine intestinal barrier (Roberts and Hill 1974). Furthermore, the intestinal epithelium of horses presents some characteristics which are uncommon in other species, e.g. the presence of conspicuous Paneth cells (Meyer et al. 1970) or granular epithelial cells. Paneth cells occur in the small intestinal crypts and sporadically in the caecum, while granular cells occur along the lining of the small and large intestine in hors...
Scintigraphic measurement of uterine clearance in normal mares and mares with recurrent endometritis.
Equine veterinary journal    March 1, 1994   Volume 26, Issue 2 109-113 doi: 10.1111/j.2042-3306.1994.tb04346.x
LeBlanc MM, Neuwirth L, Asbury AC, Tran T, Mauragis D, Klapstein E.The percentage of Technetium 99m-albumin colloid (99mTc-microAA), a radiocolloid, cleared from the uterine lumen within 4 h of intrauterine infusion, was measured in 15 mares during 2 consecutive cycles, on Day 3 of oestrus and 48 h after ovulation. Four nulliparous (Group 1) and 4 multiparous (Group 2) mares were classified as resistant and the remaining 7 multiparous mares were classified as susceptible (Group 3) to endometritis. Mares in Groups 1 and 2 cleared more 99mTc-microAA from their uteri than did mares in Group 3 during oestrus (P < 0.01) and 48 h after ovulation (P 50% of the c...
Lentivirus cross-reactive determinants present in the capsid protein of equine infectious anaemia virus.
The Journal of general virology    March 1, 1994   Volume 75 ( Pt 3) 657-662 doi: 10.1099/0022-1317-75-3-657
Grund CH, Lechman ER, Issel CJ, Montelaro RC, Rushlow KE.In this study we used immune sera from equine infectious anaemia virus (EIAV)-infected horses which uniquely display broad reactivity with different lentivirus capsid proteins (CA) to characterize the cross-reactive determinants of lentivirus CA proteins. In particular, the role of the major homology region (MHR) of lentivirus CA proteins in this serological cross-reactivity was evaluated using both equine immune serum and murine monoclonal antibodies (MAbs) directed against the MHR segment of different lentiviruses. The results of our studies indicate that about 80% of sera from long-term exp...
Identification of 2 stallion sperm-specific proteins and their autoantibody response.
Equine veterinary journal    March 1, 1994   Volume 26, Issue 2 148-151 doi: 10.1111/j.2042-3306.1994.tb04355.x
Teuscher C, Kenney RM, Cummings MR, Catten M.In this study, 2 stallions were immunised with their own spermatozoa to ascertain whether an antisperm autoantibody response could be mounted. The results demonstrated that the stallion can recognise and respond to sperm autoantigens by producing circulating antisperm antibodies, primarily of the IgG class. Such autoantibodies appeared 2-4 weeks after inoculation and persisted for 6-20 weeks. Immunochemical characterisation by western blot identified two major sperm autoantigens, with molecular weights of 70 kD and 62 kD. Control pony stallions immunised with adjuvants alone failed to exhibit ...
Experimental transmission of eastern equine encephalitis virus by strains of Aedes albopictus and A. taeniorhynchus (Diptera: Culicidae).
Journal of medical entomology    March 1, 1994   Volume 31, Issue 2 287-290 doi: 10.1093/jmedent/31.2.287
Turell MJ, Beaman JR, Neely GW.The vector competence of Aedes taeniorhynchus (Wiedemann) and four strains of Aedes albopictus (Skuse) was assessed for eastern equine encephalitis (EEE) virus isolated from Ae. albopictus collected in Polk County, Florida. Both species became infected with and transmitted EEE virus by bite after feeding on 1-d-old chicks that had been inoculated with EEE virus (viremia = 10(10.1) plaque-forming units [PFU] per ml of blood). However, when fed on an older chick with a lower viremia (viremia = 10(6.1) PFU per ml of blood), Ae. albopictus was significantly more susceptible to infection (90%, n = ...
Jugular vein wall repair after intravenous injection in equids.
Zentralblatt fur Veterinarmedizin. Reihe A    March 1, 1994   Volume 41, Issue 2 143-149 doi: 10.1111/j.1439-0442.1994.tb00077.x
Muylle S, Simoens P, Lauwers H.The evolution of the healing process of venous puncture wounds was studied macroscopically and light-microscopically in order to determine the time of venipuncture. Seven small equids of various ages, all in normal healthy condition, were experimentally injected in the external jugular vein with a physiological Hartmann solution at different times before euthanasia. Two types of needle diameters i.e. 16.5 G and 19 G, were used. A fairly good uniformity in the healing process was seen in the 7 animals. In most cases the lesions induced within 5 weeks before death were macroscopically visible, w...
Pulmonary intravascular macrophages in horses and ponies.
American journal of veterinary research    March 1, 1994   Volume 55, Issue 3 382-388
Longworth KE, Jarvis KA, Tyler WS, Steffey EP, Staub NC.Seven horses (4 anesthetized and 3 awake) and 2 ponies (anesthetized) were studied to evaluate the high sensitivity of the pulmonary circulation of the horse to various blood-borne particles, and to establish the presence of intravascular macrophages in the lung. Pulmonary and systemic pressures and cardiac output before and during particle injection were measured in some animals. An anesthetized foal had a large increase in pulmonary arterial pressure (32 and 34 mm of Hg) within 1 minute of IV administration of small test doses of radioactively labeled liposomes (2.5 mumol/kg of body weight) ...
Sperm-induced leukocytosis in the equine uterus.
Theriogenology    February 2, 1994   Volume 41, Issue 3 629-636 doi: 10.1016/0093-691x(94)90173-g
Kotilainen T, Huhtinen M, Katila T.The objective of this study was to investigate the inflammatory reaction induced in the equine uterus by insemination with fresh and frozen semen. Eleven groups (6 to 8 mares per group) were studied during 2 breeding seasons. The mares were inseminated using raw semen, frozen semen, extended fresh and frozen semen, concentrated fresh semen, seminal plasma and seminal extenders only. One group was bred naturally. Six hours after insemination, the uteri were flushed with 50 ml of phosphate-buffered saline (PBS). Seventeen out of 104 samples (16%) exhibited slight bacterial growth. Neutrophil con...
Effects of heparin, venous strangulation obstruction of the small intestine, and reperfusion of the small intestine on plasma diamine oxidase activity in horses.
American journal of veterinary research    February 1, 1994   Volume 55, Issue 2 185-191
Laws EG, Freeman DE.Diamine oxidase (DAO), an enzyme of small intestinal origin, is released from mucosal storage sites by IV administration of heparin, to yield the plasma postheparin DAO (PHD) curve. The PHD curve is diminished when mucosal surface area is lost, and baseline (without heparin) plasma DAO activity increases when mucosal storage sites are damaged. Plasma DAO activity was measured after 2 doses of heparin were administered IV in healthy, conscious horses. In anesthetized horses, the PHD curve was studied: during sham small intestinal surgery, and during venous strangulation obstruction (VSO) of the...
Production and characterization of monospecific adult worm infections of Strongylus vulgaris and Strongylus edentatus in ponies.
Veterinary parasitology    February 1, 1994   Volume 51, Issue 3-4 249-254 doi: 10.1016/0304-4017(94)90162-7
McClure JR, Chapman MR, Klei TR.Since 1978, 20 surgical implantations of either Strongylus vulgaris or Strongylus edentatus have been performed in our laboratory for the purpose of obtaining single species cultures of these parasites. Following surgical implantation peak EPG values of 13-327 (S. vulgaris) and 363-1284 (S. edentatus) generally occurred during the first 3 weeks post-implantation. Duration of infections was as long as 5 years. Successful outcome of such surgeries appears to be related to the total number of parasites used (> or = 38) and the ratio of female to male worms implanted (1:1 or 2:1).
Serum hypoxanthine and xanthine concentrations in horses heterozygous for combined immunodeficiency.
Journal of animal breeding and genetics = Zeitschrift fur Tierzuchtung und Zuchtungsbiologie    January 12, 1994   Volume 111, Issue 1-6 148-151 doi: 10.1111/j.1439-0388.1994.tb00448.x
Kettler MK, Weil MR, Mascotti K, Perryman LE.A group of diseases termed combined immunodeficiency (CID) results in a severe form of immunodeficiency. While CID in humans has two genetics bases, in Arabian it is inherited in an autosomal recessive manner. Kettler et al. (1989) determined that uric acid was significantly (p 0.05) between carrier and non-carrier horse's serum levels of hypoxanthine or xanthine. These data, combined with our previous ones suggest that an enzymatic lesion in the purine salvage pathway may occur at the urate oxidase step. Follicular fluid lipoproteins in the mare: evaluation of HDL transfer from plasma to follicular fluid.
Biochimica et biophysica acta    January 3, 1994   Volume 1210, Issue 2 226-232 doi: 10.1016/0005-2760(94)90125-2
Le Goff D.Using a density gradient ultracentrifugal procedure, we have separated equine plasma and follicular fluid high-density lipoproteins (HDL). The density distribution of the follicular fluid HDL was clearly displaced towards the highest densities in comparison with that of plasma HDL. Similarly, an analysis of size distributions showed a decrease in follicular fluid HDL diameters (4.2 to 9.2 nm) compared to plasma HDL (5.5 to 9.5 nm). HDL were isolated into three subfractions on the basis of the disposition of the Sudan Black stained bands in the centrifuge tubes. Concentrations of each subfracti...

Archiv fur Tierernahrung    January 1, 1994   Volume 46, Issue 4 367-384 doi: 10.1080/17450399409381787
Schubert R, Flachowsky G, Bochröder B.Chopped wheat straw was homogeneously mixed with urine of horses (5.75 gN per 1, 16.88 atom-% 15N-excess) and airtightly stored in plastic containers for 6 months. Three rumen fistulated sheep and goats each were fed with untreated or urine treated straw. Concentrate was added to straw. Untreated and urine treated straw were given in nylon bags and incubated in the rumen of sheep and goats for 1, 3, 6, 12, 24, 48 and 72 hours. A three compartment exponential function was used to fit the measurements of 15N-excess and 15N-amount of bag content. The curves and the calculated partial Y-values of ...
Measurements of urethral pressure profiles in the male horse.
Equine veterinary journal    January 1, 1994   Volume 26, Issue 1 55-58 doi: 10.1111/j.2042-3306.1994.tb04332.x
Ronen N.Urinary tract pressure profiles were evaluated in 6 clinically normal geldings over 3 consecutive days. This was performed by introducing a 1.3 m-long cuffed catheter into the urinary tract, under xylazine sedation (0.8 mg/kg, iv). The method was reproducible. The mean (+/- sd) intra-vesicular pressure (IVP) and maximal urethral closure pressures (MUCP) were 10.3 (+/- 1.7) and 129.8 (+/- 19.6) cmH2O, respectively, and the ratio between MUCP and IVP was 13.2 (+/- 2.5). A gelding with urinary incontinence showed a significantly lower MUCP (73.4 cmH2O), and an MUCP to IVP ratio of 8.0. It was con...

Natural outcome and ultrasonic identification of equine fetal twins.
Theriogenology    January 1, 1994   Volume 41, Issue 5 1193-1199 doi: 10.1016/s0093-691x(05)80041-x
Ginther OJ, Griffin PG.The natural outcome of bilateral twins (one in each uterine horn) that were viable on Day 40 was studied in 15 pony mares. Fetuses were monitored by transrectal ultrasonography until the outcome was determinable. One fetus (2 mares) or both fetuses (8 mares) died (cessation of heartbeats) during Months 2 and 3. Loss of fetuses was externally observed in 4 additional mares during the late fetal stage (Months 8 to 11); 2 mares lost 1 fetus and 2 lost both fetuses. Birth of 2 live foals occurred in only 1 of 15 mares (7%) First day of detected apposition between portions of the 2 allantochorions ...
Insemination results with slow-cooled stallion semen stored for approximately 40 hours.
Acta veterinaria Scandinavica    January 1, 1994   Volume 35, Issue 3 257-262 doi: 10.1186/BF03548330
Heiskanen ML, Huhtinen M, Pirhonen A, Mäenpää PH.Semen from 3 stallions was extended using 2 methods (Kenney extender and a modified Kenney extender), slowly cooled, and stored for 41 +/- 6 (s.d.) h before insemination. An insemination dose (40 ml) contained 1.5-2 billion spermatozoa. In the experiment, 26 mares were inseminated in 30 cycles. The pregnancy rate per cycle obtained with sperm stored in the Kenney extender was 87% (n = 15). When the semen was extended with the modified extender, centrifuged and stored, the pregnancy rate was 60% (n = 15). Inseminations were done every other day until ovulation was detected. If a mare ovulated m...
The development of Babesia (Theileria) equi (Laveran, 1901) in the gut and the haemolymph of the vector ticks, Hyalomma species.
Parasitology research    January 1, 1994   Volume 80, Issue 4 297-302 doi: 10.1007/BF02351869
Zapf F, Schein E.The development of the piroplasm Babesia equi was studied by light microscopy in the gut and the haemolymph of three different Hyalomma species during and after the nymphs had engorged on parasitaemic horses. The stock of B. equi used was isolated from a horse imported from Turkmenistan (CIS) in 1991. The existence of gamogony was identified by the occurrence of gamonts and gametes in the gut contents of the nymphs at between 3 and 4 days after infestation of the nymphs, before the ticks dropped off the experimentally infected horses. Zygotes and kinetes were observed in the intestinal cells f...
Pregnancies following transfer of equine embryos cryopreserved by vitrification.
Theriogenology    January 1, 1994   Volume 42, Issue 3 483-488 doi: 10.1016/0093-691x(94)90686-d
Hochi S, Fujimoto T, Braun J, Oguri N.The objective of this study was to investigate the in vitro and in vivo developmental abilities of equine embryos cryopreserved by vitrification. Twenty-eight embryos were recovered from Native pony and Thoroughbred mares at Days 5 to 7 by nonsurgical uterine flushing (detection of ovulation=Day 0). The vitrification solution contained 40% ethylene glycol, 18% Ficoll, and 0.3 M sucrose in PBS. The embryos were placed for 1 to 2 min in vitrification solution (Group 1) or following exposure to 20% ethylene glycol in PBS for 10 to 20 min (Groups 2 and 3). Single embryos were loaded in 0.25-ml str...
Inhibitory nerve distribution and mediation of NANC relaxation by nitric oxide in horse airways.
Journal of applied physiology (Bethesda, Md. : 1985)    January 1, 1994   Volume 76, Issue 1 339-344 doi: 10.1152/jappl.1994.76.1.339
Yu M, Wang Z, Robinson NE, Leblanc PH.The distribution of inhibitory nerves and the mediator of the inhibitory nonadrenergic noncholinergic (iN-ANC) nervous system were investigated in smooth muscle preparations from seven regions of equine airways. In tissues incubated with atropine and precontracted with histamine, electrical field stimulation produced frequency-dependent relaxation, and the magnitude of the relaxation decreased from trachea to central bronchi and was absent in peripheral airways. The degree of relaxation in bronchi was not simply a function of bronchial size or generation. Propranolol inhibited part of the rela...
Inhibin activity in the mare and stallion.
Domestic animal endocrinology    January 1, 1994   Volume 11, Issue 1 87-100 doi: 10.1016/0739-7240(94)90037-x
Roser JF, McCue PM, Hoye E.An overnight double antibody RIA, employing a rabbit antiserum raised to bovine 31 kDa inhibin (rAs-#1989, NICHD) and purified bovine 31 kDa inhibin (bINH-I-90/1, NICHD) as trace and standard, was validated to measure immunoreactive inhibin (iINH) concentrations in equine peripheral plasma, follicular fluid (FF), ovarian vein (OV) plasma, testicular tissue extracts (TTE) and testicular vein (TV) plasma. The dynamic relationship of iINH and follicle stimulating hormone (FSH) was investigated during the estrous cycle of the mare and the annual reproductive cycle of the stallion. In the RIA, para...
