Phagocytosis and intracellular killing of the contagious equine metritis organism by equine neutrophils in genital secretions. Equine neutrophils were combined with contagious equine metritis organism (CEMO) or Escherichia coli in vitro in the presence of seminal plasma, uterine flushings, or Hank's balanced salt solution (HBSS). Phagocytosis and intracellular killing were estimated by bacterial culture and light and electron microscopy. With lysed neutrophils, the numbers of colony-forming units of CEMO and E coli increased in seminal plasma and uterine flushings. Numbers of CEMO decreased in HBSS. Ultrastructure of the equine hoof wall secondary epidermal lamellae. The ultrastructure of cells of the secondary epidermal lamellae from the hoof wall of 8 adult horses was described. Stages of progressive keratinization were not observed in this cell population. The morphologic features of cells of the secondary epidermal lamellae indicated that these lamellae were composed of nonmobile populations of cells and that primary epidermal lamellae moved past the secondary lamellae by breaking of the desmosomes connecting these 2 cell populations. Fecal leukocytes and epithelial cells in horses with diarrhea. Fecal samples from 74 horses with diarrhea were examined microscopically for leukocytes and epithelial cells. The diarrhea was categorized as either acute (less than two weeks) or chronic, Salmonella positive or negative, and mild or severe (based on the fecal consistency). A large number of fecal leukocytes was strongly suggestive of salmonellosis; however, approximately 1/3 of the horses with Salmonella-negative diarrhea also shed fecal leukocytes. Detection of rotavirus in horses with and without diarrhea by electron microscopy and Rotazyme test. A total of 142 equine fecal samples (93 field fecal and 49 experimental fecal specimens) were examined for rotavirus using direct electron microscopy (EM) and the Rotazyme test. Eighty-six stool specimens were diarrhea samples. The Rotazyme test sensitivity and accuracy as compared to EM was determined by the visual (color reaction) and spectrophotometric methods. Malignant melanomas in farm animals. Malignant melanomas in 5 horses, 1 calf, 1 cow and 2 pigs were described. The disease, in white or gray horses, was tegumentally located and generalized in the organs of the abdominal cavity. Microscopically, dendritic and fusiform pigment cells and multinucleated cells were identified. In the hypophysis there microcysts and in one case a hypophyseal adenoma were encountered. The pathology of experimental Corynebacterium equi infection in foals following intragastric challenge. The intragastric inoculation of a suspension of Corynebacterium equi on five consecutive days induced severe ulcerative colitis, typhlitis, and lymphadenitis of colonic and cecal nodes in two ponies necropsied three weeks after infection. No gross lesions were observed in two ponies necropsied ten days after infection. A single inoculum of equivalent size failed to induce gross lesions in four ponies killed at ten or 20 days after infection. The isolation and preliminary characterization of a rhabdovirus in Australia related to bovine ephemeral fever virus. CSIRO 368 virus was isolated from blood collected in the Northern Territory from a healthy cow and electron microscope studies showed that the isolate had rhabdovirus morphology. Fluorescent antibody studies and complement fixation tests related the virus to bovine ephemeral fever (BEF) virus. Neutralization tests in both suckling mice and Vero cells showed that the virus was not BEF virus. Structural and functional organization of the suprapatella in two cercopithecines. Gross and microscopic study of Cercopithecus aethiops pygerythrus and Papio cyanocephalus anubis shows that these cercopithecines have a quadriceps tendon the distal portion of which consists mostly of dense collagenous bundles with scattered fine elastic fibres most of which lie in the loose connective tissue planes within and around the tendon and around blood vessels. A morphological study of the lesions of African horsesickness. Gross, histological and ultrastructural findings are described in 6 natural cases and in 2 experimental cases of African horsesickness. From the gross lesions the cases were divisible into 2 groups which represented the previously described pulmonary and mixed forms of the disease. Fine structure of Babesia equi Laveran, 1901 within lymphocytes and erythrocytes of horses: an in vivo and in vitro study. The development of Babesia equi (Piroplasmia) in the vertebrate host was studied by electron microscopy. The tick-borne sporozoites initiated an exoerythrocytic schizogony in lymphocytes in vivo and in vitro. Polymorphonuclear neutrophil leucocytes of peritoneal fluid. Cells in the peritoneal fluid from 179 horses were examined in Giemsa stained preparations using light microscopy. Neutrophils were found in all samples whether transudative or exudative although their proportions varied enormously. The venous organisation of the dermal lamellae of the equine hoof wall of 18 pairs of forelimb hooves from clinically healthy horses was studied using plastic vascular corrosion casts, and scanning electron microscopic and histological techniques. Congenital intestinal aganglionosis in white foals. A congenital and probably hereditary neurological defect has been identified in the intestinal tract of six foals produced from the breeding of overo (a type of spotting pattern) horses. The foals had white hair and pink skin with the exception of occasional pigmented foci about the muzzle, ventral abdomen, and hindquarters. Annular gap junctions of the equine hoof wall. Incidental to studies of keratinization of the equine hoof wall, annular gap junctions were found in the stratum spinosum of the intertubular horn of the stratum medium. Adjacent cells of the stratum spinosum showed extensive gap junctions, and often local invaginations of one cell into another were bound by gap junctions. The uptake of mepacrine by horse polymorphonuclear leucocytes in vitro. The uptake of mepacrine by isolated horse polymorphonuclear leucocytes (PMN) was measured using spectrophotofluorimetry. Two phases of uptake were observed, the first, rapid fraction, essentially complete by 10 min, and a second, slow fraction, which was still proceeding after 60 min. In uterine or cervical specimens obtained from pony mares infected with streptomycin-resistant contagious equine metritis bacteria, several colonies of the bacteria which differed in morphologic characteristics were recognized during their primary isolation on Eugon chocolate agar and tryptose chocolate agar plates. Mesothelial cells of peritoneal fluid. Cells in the peritoneal fluid from 159 horses were examined in Giemsa stained preparations using light microscopy. Normal mesothelial cells showed an oval nucleus with finely reticular chromatin and pale blue cytoplasm. Activated mesothelial cells occurred in fluids derived from mesothelium under acute or subacute, non-septic stimulus and were remarkable for their pleomorphism and intense basophilia which may mimic neoplasia. Transformed mesothelial cells seen in chronic inflammatory fluids were sometimes phagocytic and showed conspicuous cytoplasmic vacuolation.
Transmission electron microscopy of horse embryos 3-16 days after ovulation. The 23 embryos were obtained by flushing the reproductive tract. Though the general cytology was observed, most attention was given to the formation of the embryonic capsule. It first appeared as a thin uniform layer on the inner surface of the zona pellucida of embryos recovered from the uterus on Day 6. By Day 8 the capsule was about 1 micron thick and the zona pellucida had been shed.
