Topic:Leukocytes

Leukocytes, also known as white blood cells, are an integral component of the equine immune system, responsible for defending horses against infections and foreign substances. These cells are produced in the bone marrow and circulate throughout the bloodstream and tissues. In horses, leukocytes are categorized into several types, including neutrophils, lymphocytes, monocytes, eosinophils, and basophils, each with specific functions in immune response. Neutrophils are involved in phagocytosis and the destruction of pathogens, while lymphocytes are key players in adaptive immunity. Monocytes differentiate into macrophages and dendritic cells, contributing to pathogen clearance and antigen presentation. Eosinophils and basophils are associated with allergic reactions and parasitic infections. Variations in leukocyte counts and distribution can indicate underlying health conditions, making them valuable in veterinary diagnostics. This page aggregates peer-reviewed research studies and scholarly articles that explore the physiology, function, and diagnostic relevance of leukocytes in equine health.
Detection and comparison of nitric oxide in clinically normal horses and those with naturally acquired small intestinal strangulation obstruction.
Canadian journal of veterinary research = Revue canadienne de recherche veterinaire    October 26, 1999   Volume 63, Issue 4 230-240
Mirza MH, Oliver JL, Seahorn TL, Hosgood G, Moore RM.The purpose of this study was to determine whether nitric oxide (NO) is present in clinically normal horses under basal conditions and if it increases secondary to naturally acquired small intestinal strangulation obstruction. Thirty-one horses were used; 20 horses with naturally acquired small intestinal strangulation obstruction and 11 clinically normal horses with no signs of gastrointestinal tract disease. Jugular venous blood, abdominal fluid, and urine were collected for NO quantification. Plasma, abdominal fluid, and urine were stored at -70 degrees C until analyzed for NO using a chemi...

Novel cathelicidins in horse leukocytes(1).
FEBS letters    September 3, 1999   Volume 457, Issue 3 459-464 doi: 10.1016/s0014-5793(99)01097-2
Scocchi M, Bontempo D, Boscolo S, Tomasinsig L, Giulotto E, Zanetti M.Cathelicidins are precursors of defense peptides of the innate immunity and are widespread in mammals. Their structure comprises a conserved prepropiece and an antimicrobial domain that is structurally varied both intra- and inter-species. We investigated the complexity of the cathelicidin family in horse by a reverse transcription-PCR-based cloning strategy of myeloid mRNA and by Southern and Western analyses. Three novel cathelicidin sequences were deduced from bone marrow mRNA and designated equine cathelicidins eCATH-1, eCATH-2 and eCATH-3. Putative antimicrobial domains of 26, 27 and 40 r...
The role of prostanoids and nitric oxide in endotoxin-induced hyporesponsiveness of equine digital blood vessels.
Equine veterinary journal    July 13, 1999   Volume 31, Issue 3 212-218 doi: 10.1111/j.2042-3306.1999.tb03175.x
Bailey SR, Elliott J.Endotoxin has been implicated in the pathophysiology of acute laminitis. The aim of this study was to examine the direct effects of endotoxin on isolated equine digital blood vessels. Equine digital veins (EDV), incubated in Krebs-Henseleit solution containing lipopolysaccharide (LPS) (1 microg/ml) became hyporesponsive to 5-HT after 16 h. Cycloheximide and ibuprofen blocked this effect of LPS and increased the maximum response obtained to 5-HT when compared to control vessels. L-nitroarginine methyl ester (L-NAME) reversed the hyporesponsiveness caused by LPS. Vessels maintained in culture me...
A pharmacodynamic and pharmacokinetic study with vedaprofen in an equine model of acute nonimmune inflammation.
Journal of veterinary pharmacology and therapeutics    June 18, 1999   Volume 22, Issue 2 96-106 doi: 10.1046/j.1365-2885.1999.00173.x
Lees P, May SA, Hoeijmakers M, Coert A, Rens PV.The pharmacodynamics and enantioselective pharmacokinetics of vedaprofen were studied in six ponies in a two period cross-over study, in which a mild acute inflammatory reaction was induced by carrageenan soaked sponges implanted subcutaneously in the neck. Vedaprofen, administered intravenously at a dosage of 1 mg/kg, produced significant and prolonged inhibition of ex vivo serum thromboxane B2 (TXB2) synthesis and short-lived inhibition of exudate prostaglandin E2 (PGE2) and TXB2 synthesis. Vedaprofen also partially inhibited oedematous swelling and leucocyte infiltration into exudate. Vedap...
Plasma myeloperoxidase level and polymorphonuclear leukocyte activation in horses suffering from large intestinal obstruction requiring surgery: preliminary results. Grulke S, Benbarek H, Caudron I, Deby-Dupont G, Mathy-Hartert M, Farnir F, Deby C, Lamy M, Serteyn D.Myeloperoxidase (MPO) is a specific enzyme of neutrophil azurophilic granules with a strong oxidative activity. Thanks to a radioimmunoassay of equine myeloperoxidase, the authors have observed a significantly higher plasma level of MPO in horses operated for strangulation obstruction of the large intestine (n = 6) than in horses suffering from a non-strangulating displacement of the large intestine (n = 9). For the 2 groups, 3 phases were distinguished: reception (P1), intensive care (P2) and terminal phase (P3). The mean peak values of MPO for these phases were 121.6 ng/mL (P1), 168.6 ng/mL ...

Journal of comparative pathology    March 31, 1999   Volume 120, Issue 1 1-13 doi: 10.1053/jcpa.1998.0250
Banks EM, Kyriakidou M, Little S, Hamblin AS.Leucocytes in the lung epithelium play an important role in the ability of an animal to respond appropriately to inhaled pathogens. The distribution of lymphoid and myeloid cells associated with the lung epithelium was examined immunohistochemically throughout the respiratory tract of four horses, comprising two adults from an abattoir, one pregnant mare, and her fetus (in the final stage of gestation). Cross and tangential cryosections were labelled with monoclonal antibodies against T-cell, B-cell, macrophage/dendritic myeloid cell, and major histocompatibility Class (MHC) II surface antigen...
In vitro reactivation of latent equid herpesvirus-1 from CD5+/CD8+ leukocytes indirectly by IL-2 or chorionic gonadotrophin.
The Journal of general virology    January 8, 1999   Volume 79 ( Pt 12) 2997-3004 doi: 10.1099/0022-1317-79-12-2997
Smith DJ, Iqbal J, Purewal A, Hamblin AS, Edington N.IL-2 and equine chorionic gonadotrophin (eCG) initiated reactivation of equid herpesvirus-1 (EHV-1) from venous lymphocytes at a frequency of 1/10(-5). Indirect immunofluorescence showed that > 80% of virus-positive leukocytes were CD5+/CD8+ with the remaining 20% being CD5+/CD8-/CD4-. Cocultivation demonstrated that the reactivated virus was infectious. In addition, virus was reactivated in vitro from leukocytes of > 70% of horses by the mitogens phytohaemagglutinin (PHA) and pokeweed mitogen (PWM). Transfer of supernatants showed that IL-2 and eCG acted indirectly by causing the releas...
Flow cytometric determination of oxidative burst activity of equine peripheral blood and bronchoalveolar lavage-derived leucocytes.
Veterinary journal (London, England : 1997)    November 7, 1998   Volume 156, Issue 2 117-126 doi: 10.1016/s1090-0233(05)80037-1
Raidal SL, Bailey GD, Love DN.Flow cytometric techniques were developed for the evaluation of oxidative burst activity in equine peripheral blood neutrophils and lymphocytes, as well as bronchoalveolar lavage derived pulmonary alveolar macrophages and lymphocytes. The oxidation of dichlorofluorescin was measured by the increased fluorescence of cells stimulated with phorbol myristate acetate or a variety of other stimulants. Flow cytometry was a suitable method for the evaluation of the intracellular oxidation in all cell populations evaluated. Analysis was rapid and cell separation before analysis was not required. Hetero...
The flow cytometric evaluation of phagocytosis by equine peripheral blood neutrophils and pulmonary alveolar macrophages.
Veterinary journal (London, England : 1997)    November 7, 1998   Volume 156, Issue 2 107-116 doi: 10.1016/s1090-0233(05)80036-x
Raidal SL, Bailey GD, Love DN.Flow cytometry was used to assess the phagocytosis of fluorescent-labelled bacteria by equine peripheral blood neutrophils and pulmonary alveolar macrophages. Cell populations were prepared from venous blood following ammonium chloride lysis and from washed bronchoalveolar lavage derived samples. Discrete clusters of cells, corresponding to different leucocyte groups, were readily identified on the basis of differing light scattering properties and could thus be discriminated, negating the need for prior cell separation. Cells able to associate with fluorescent-labelled bacteria (by attachment...
Experimental infection of four horses with Ehrlichia phagocytophila.
The Veterinary record    October 28, 1998   Volume 143, Issue 11 303-305 doi: 10.1136/vr.143.11.303
Pusterla N, Lutz H, Braun U.Four clinically healthy horses which were negative for antibodies to Ehrlichia phagocytophila, the agent of bovine ehrlichiosis, were infected experimentally with E phagocytophila-containing bovine leucocytes, administered intravenously. The horses were examined daily for four weeks, and blood samples were collected daily for cytological, haematological and biochemical examination and for a nested polymerase chain reaction (PCR). An indirect immunofluorescence test was used to determine when the horses seroconverted and the duration of positive titres. There were no abnormal clinical, haematol...
A comparative study of mast cells and eosinophil leukocytes in the mammalian testis.
Zentralblatt fur Veterinarmedizin. Reihe A    August 11, 1998   Volume 45, Issue 4 209-218 doi: 10.1111/j.1439-0442.1998.tb00819.x
Anton F, Morales C, Aguilar R, Bellido C, Aguilar E, Gaytán F.The existence of a physiological integration between the immune and endocrine systems has long been recognized. In spite of the abundant literature data on the presence of cells of the immune system in the testis, mast cells and eosinophil leukocytes have received little attention. We have studied the presence, distribution and numbers of mast cells and eosinophils in the testes of 12 mammalian species. Mast cells were frequently found in equine (stallion, ass and mule) and human testis, whereas eosinophils were nearly absent. On the contrary, eosinophils were abundant in the hare testis, whil...
Inhaled leukotrienes cause bronchoconstriction and neutrophil accumulation in horses.
Research in veterinary science    August 5, 1998   Volume 64, Issue 3 219-224 doi: 10.1016/s0034-5288(98)90129-1
Marr KA, Lees P, Page CP, Cunningham FM.Leukotrienes have been shown to mimic many of the pathophysiological processes in allergic airway disease. In this study the bronchoconstrictor effect of inhaled LTD4, and radiolabelled neutrophil accumulation in response to inhalation of LTB4, have been examined in the horse. In separate studies, solutions of LTD4 and LTB4 were administered to the airways of normal animals by nebulisation. LTD4, but not LTB4, caused a dose-dependent increase in pleural pressure which was maximal at three to four minutes and had returned to baseline by 15 to 20 minutes. On a molar basis LTD4 was 305 to 970 tim...
Effect of the 5-lipoxygenase inhibitor, fenleuton, on antigen-induced neutrophil accumulation and lung function changes in horses with chronic obstructive pulmonary disease.
Journal of veterinary pharmacology and therapeutics    July 23, 1998   Volume 21, Issue 3 241-246 doi: 10.1046/j.1365-2885.1998.00127.x
Marr KA, Lees P, Page CP, Cunningham FM.The leukotrienes (LT) LTD4 and LTB4 have been shown to cause bronchoconstriction and neutrophil accumulation, respectively, in horse lungs. Such changes are characteristic of the equine allergic respiratory disease, chronic obstructive pulmonary disease (COPD). To further investigate the role of these putative mediators in the pathogenesis of equine COPD the effect of a 5-lipoxygenase inhibitor, fenleuton, on antigen-induced changes in horses with this condition has been examined. Six horses with COPD underwent a series of four antigen challenges, one month apart, with placebo pre-treatment on...

Research in veterinary science    June 13, 1998   Volume 64, Issue 2 111-117 doi: 10.1016/s0034-5288(98)90005-4
Marr K, Marsh K, Hernandez L, Cunningham FM, Lees P.Leukotrienes, products of the 5-lipoxygenase pathway of arachidonic acid metabolism, possess properties consistent with their involvement in a range of inflammatory diseases. In this study the pharmacokinetics and pharmacodynamics of the selective 5-lipoxygenase inhibitor, fenleuton, have been examined in the horse. Orally administered fenleuton (four 5 mg kg(-1) doses, given once daily) was absorbed from the gastrointestinal tract, and penetrated readily into tissue cage exudate, the ratio of the plasma:exudate AUC0-48h being 0.90+/-0.02 (n=6). Ionophore-stimulated leukotriene (LT) B4 synthes...
A dinucleotide mutation in the endothelin-B receptor gene is associated with lethal white foal syndrome (LWFS); a horse variant of Hirschsprung disease.
Human molecular genetics    June 13, 1998   Volume 7, Issue 6 1047-1052 doi: 10.1093/hmg/7.6.1047
Yang GC, Croaker D, Zhang AL, Manglick P, Cartmill T, Cass D.Lethal white foal syndrome (LWFS) is a congenital anomaly of horses characterized by a white coat colour and aganglionosis of the bowel, which is similar to Hirschsprung disease (HSCR). We decided to investigate possible mutations of the endothelin-B receptor gene ( EDNRB ) in LWFS as recent studies in mutant rodents and some patients have demonstrated EDNRB defects. First, we identified a full-length cDNA for horse EDNRB . This cDNA fragment contained a 1329 bp open reading frame which encoded 443 amino acid residues. The predicted amino acid sequence was 89, 91 and 85% identical to human, bo...
Isolation and characterisation of equine dendritic cells.
Veterinary immunology and immunopathology    April 9, 1998   Volume 60, Issue 1-2 15-31 doi: 10.1016/s0165-2427(97)00093-7
Siedek E, Little S, Mayall S, Edington N, Hamblin A.Despite their important role in initiating T-cell responses in other species, dendritic cells have not been studied in the horse. A method for isolating blood dendritic cells by adherence and metrizamide gradients was adapted to equine cells. A number of monoclonal antibodies (mAbs), including some which label dendritic cells in other species, were tested for immunochemical reactivity with the isolated blood dendritic cells, and sections of lymph node and spleen. 62 +/- 6% of the isolated blood cells were MHC Class II positive and had typical dendritic cell morphology and only 4 +/- 2% contain...
Failure of lipopolysaccharides to directly trigger the chemiluminescence response of isolated equine polymorphonuclear leukocytes.
Veterinary research communications    November 5, 1997   Volume 21, Issue 7 477-482 doi: 10.1023/a:1005938319482
Benbarek H, Deby-Dupont G, Caudron I, Deby C, Lamy M, Serteyn D.Divergent results have been reported on the effects of lipopolysaccharides (LPS) on the activation of equine polymorphonuclear leukocytes (PMN). We therefore attempted to determine whether LPS alone can stimulate equine PMN or whether plasma factors are necessary. PMN were isolated from citrated blood on a discontinuous density gradient of Percoll. The luminol (10(-3) mol/L)-enhanced chemiluminescence (CL) of 1.25 x 10(6) cells was measured after addition of Escherichia coli LPS (0.001-10 micrograms/ml) alone or after incubation in autologous plasma (1 h, 37 degrees C). After direct stimulatio...

Journal of veterinary pharmacology and therapeutics    August 1, 1997   Volume 20, Issue 4 296-307 doi: 10.1046/j.1365-2885.1997.00064.x
Cunningham FM, Andrews M, Landoni MF, Lees P.Calcium ionophore A23187 induced time and concentration dependent production of immunoreactive leukotriene (LT) B4 by equine heparinized whole blood in vitro. Time dependent production of immunoreactive LTB4 by equine neutrophils and immunoreactive LTC4 by equine eosinophils in vitro was also demonstrated. The 5-lipoxygenase activating protein (FLAP) inhibitors, BAY X 1005 and BAY Y 1015, produced concentration dependent inhibition of ionophore-induced LTB4 synthesis by equine whole blood (mean +/- SEM IC50s n = 5; 6.14 +/- 0.28 microM vs. 12.30 +/- 0.75 microM for BAY Y 1015 and BAY X 1005, r...
Cell-mediated cytolysis of equine herpesvirus-infected cells by leukocytes from young vaccinated horses.
Veterinary immunology and immunopathology    July 1, 1997   Volume 57, Issue 3-4 201-214 doi: 10.1016/s0165-2427(96)05749-2
Ellis JA, Steeves E, Wright AK, Bogdan JR, Davis WC, Kanara EW, Haines DM.The objective of this study was to determine whether the administration of modified-live equine herpesvirus (EHV-1) to young horses with residual maternal antibodies stimulated EHV-specific cytolytic responses, and whether these responses were crossreactive between EHV-1 and EHV-4. Eighteen clinically normal Belgian cross-foals were used in the study and were commingled in two adjacent pens. Skin biopsies were harvested from 16 foals within 24 h of birth and fibroblast cultures were established, expanded and cryopreserved. Beginning at approximately 10 weeks of age, 10 randomly chosen foals we...
Acute myelogenous leukaemia in a mare.
Australian veterinary journal    May 1, 1997   Volume 75, Issue 5 329-331 doi: 10.1111/j.1751-0813.1997.tb15702.x
Ringger NC, Edens L, Bain P, Raskin RE, Larock R.A 5-year-old Thoroughbred mare presented with a 4 week history of weight loss, fever and leukopenia. Rectally, a large active foetus, thickened spleen and an abdominal mass were palpated. Leukopenia, mild anaemia, marked thrombocytopenia and hyperfibrinogenaemia were found. Cytology and cytochemical staining of a bone marrow aspirate supported a diagnosis of acute myelogenous leukaemia. The mare deteriorated despite medical therapy and was humanely euthanased.
Detection of latency-associated transcripts of equid herpesvirus 1 in equine leukocytes but not in trigeminal ganglia.
Journal of virology    May 1, 1997   Volume 71, Issue 5 3437-3443 doi: 10.1128/JVI.71.5.3437-3443.1997
Chesters PM, Allsop R, Purewal A, Edington N.Results from Southern hybridization and PCR amplification experiments using a randomly synthesized reverse transcription-PCR product showed that peripheral blood leukocytes from horses showing no clinical signs of disease expressed a putative latency-associated transcript antisense to and overlapping the 3' end of the equid herpesvirus 1 (EHV-1) immediate-early gene (gene 64). A PCR product derived from this transcript has > or =96% identity with the published EHV-1 sequence. In situ hybridization studies of equine bronchial lymph nodes corroborated these findings and are consistent with re...
