HOLTON FA.In 1941, Keilin & Mann described an optical method of studying the penetration of erythrocytes by bicarbonate ions and the intracellular activity of carbonic anhydrase. This method consists of converting the haemoglobin inside the cell to methaemoglobin and observing the transformation of the methaemoglobin from the acid to the alkaline form by the entering bicarbonate ions. During this work they noticed that this transformation did not occur if the methaemoglobin cells had been washed in acid phosphate and interpreted this as an effect on the permeability of the cells.
