The immune response in horses involves a complex network of cells, tissues, and molecules that work together to protect the animal from pathogens and other harmful agents. This process includes both innate and adaptive immune mechanisms. Innate immunity provides the first line of defense and involves components such as physical barriers, phagocytic cells, and the complement system. Adaptive immunity, on the other hand, is characterized by the activation of lymphocytes and the production of antibodies, which provide a targeted response to specific antigens. Key components of the equine immune system include T cells, B cells, and various cytokines that facilitate communication between immune cells. This page compiles peer-reviewed research studies and scholarly articles that explore the mechanisms, regulation, and implications of immune responses in equine health.
Hietala SK, Ardans AA, Sansome A.An enzyme-linked immunosorbant assay was developed to measure naturally occurring Corynebacterium equi specific antibody in horse serum. Antibody against C equi was demonstrated in normal adults and was passively transferred to foals. Adult levels of specific antibody were reached by 5 to 6 months of age in healthy foals. Decreased early antibody levels were demonstrated in a limited number of foals with confirmed C equi infection.
Bertram TA.Differences in neutrophil morphology between various species of domestic animals are evident when morphometric techniques are used. Morphometric analysis can be coupled with functional assays of degranulation to demonstrate changes in granule volume after neutrophil activation (Bertram and Jensen, 1984). Morphometric and functional analysis of the neutrophil can also be used to evaluate the response of neutrophils to infectious agents (Coignoul et al., 1984a). Comparison of these assays between animal species may provide insight into the susceptibility of animals to various microbial pathogens...
Harkiss GD.An isotype-specific microELISA is presented for the measurement of antibodies to equine antithymocyte globulin in human heart transplant recipients. The assay conditions were optimized and evaluated in serial samples from 40 patients receiving a cardiac allograft. The results demonstrate that despite steroid immunosuppression and T cell cytopenia the majority of patients receiving antithymocyte globulin develop significant antibody responses, with some producing very high titres. IgM and IgG isotypes tended to predominate, with peak antibody responses occurring during the second and third week...
Ellenberger MA, Kaeberle ML, Roth JA.A lymphocyte blastogenic assay was developed to serve as an in vitro correlate of cell-mediated immunity to Rhodococcus (Corynebacterium) equi (R equi) in the equine species. Lymphocytes obtained from a group of experimental ponies showed no response in cell culture to R equi heat extract or lysozyme extract antigens. Ponies were assigned to groups for experimental inoculation. Three ponies were inoculated subcutaneously with live R equi, 3 were given live R equi by intranasal and intratracheal routes, and 4 ponies were left untreated. Lymphocytes from all inoculated ponies had a mitogenic res...
Hines MT.The continued study of immunology and its relationship to diseases of the eye will hopefully give some insight into the pathogenic mechanisms of certain ocular diseases of many species, including the horse. It may lead to a better understanding of equine recurrent uveitis, a disease that has remained an enigma for years and that now appears to be an immunologic hypersensitivity response to a number of varied antigens. The precise mechanism of the inflammation is still unclear, and the immunologic response may be variable or mixed depending upon the inciting antigen. Other ophthalmic diseases i...
Coignoul FL, Bertram TA, Cheville NF.Neutrophils isolated from venous blood of adult and foal ponies inoculated with equine herpesvirus-1 were evaluated by in vitro function tests and by electron microscopy. Foals had fever and severe neutropenia 24 hours after inoculation; increased neutrophil random migration under agarose and decreased antibody-dependent cell-mediated cytotoxicity were significant at 24 hours, but values had returned to preinoculation levels by 72 hours. Mares had fever and leukopenia of less severity, increases in neutrophil migration, and longer persistence of primary granule release than were seen in foals....
Fitzpatrick DR, Studdert MJ.The specificity of selected immune responses to equine herpesvirus type 1 (EHV-1) and type 4 (EHV-4) was examined in 3 colostrum-deprived specific-pathogen-free foals. Single foals were vaccinated with inactivated EHV-1, inactivated EHV-4, or control cell lysate plus adjuvant followed by successive intranasal challenge exposures with EHV-1 and EHV-4 or with EHV-4 and EHV-1. Vaccination with inactivated virus preparations elicited cellular immune responses and antibody which were augmented by subsequent challenge exposures. Cellular immune responses, as measured by in vitro lymphocyte blastogen...
Montelaro RC, Parekh B, Orrego A, Issel CJ.The recurrent nature of equine infectious anemia has been attributed to relatively rapid antigenic variations in equine infectious anemia virus (EIAV) during persistent infection under selective immune pressures. This model was tested by serological and biochemical analysis of virus isolates recovered from separate febrile episodes in two experimentally infected ponies. Neutralization assays employing immune sera from the experimentally infected ponies demonstrated that distinct antigenic strains of virus predominate during sequential febrile episodes in a single pony. Analysis of the test str...
The importance of the horse industry to the economy of the United States and the impact of parasitic infections on the industry are well documented. However, contemporary research activity on internal parasites of horses has not kept pace with growth of the horse population. Parasitic infections are a major facet of enteritis and colic in horses. Parasites are also associated with poor growth and development, respiratory tract disease, dermatitis, and CNS lesions. Babesia infections remain a threat to horses imported from some regions of the world. Most research activity has dealt with the dev...
Payne S, Parekh B, Montelaro RC, Issel CJ.The unique periodic nature of equine infectious anaemia (EIA) is believed to result from the ability of the infecting virus. EIAV, to undergo relatively rapid antigenic variations which circumvent host immune responses resulting in distinct virus populations in sequential clinical episodes in the persistently infected horse. This model was examined by oligonucleotide mapping comparisons of the RNA genomes of selected isolates of EIAV. Variations in oligonucleotide maps could be reproducibly demonstrated (i) after adaptation of the laboratory strain of EIAV to replication in a pony, (ii) after ...
Antczak DF, Miller JM, Remick LH.Evidence is presented for a reproducible maternal immune response to histocompatibility antigens during equine pregnancy. Mares were stimulated as a result of pregnancy to produce cytotoxic antibodies to paternal lymphocyte alloantigens. The majority of these antibodies were directed against antigens of the equine lymphocyte antigen (ELA) system, which is the major histocompatibility complex (MHC) of the horse. In 16 experimental pregnancies produced using 12 mares and 4 stallions which had been typed for ELA antigens, there was correlation between ELA incompatibility between sire and dam and ...
Montelaro RC, West M, Issel CJ.The immunogenic contributions of the carbohydrate and peptide portions of the major envelope glycoprotein of equine infections anemia virus, EIAV gp90, were analyzed by measuring the effects of specific glycosidase and protease digestions on the reactivity of the glycoprotein with immune sera from infected horses. The results of both direct and competitive radioimmunoassay demonstrated that immune sera contained antibodies reactive with both the carbohydrate and protein moieties of EIAV gp90, with the predominant reactivity apparently against the gp90 peptide epitopes. These results contrast w...
Mumford JA, Bates J.Serological responses following two and three doses of an inactivated equid herpesvirus 1 ( EHV -1) vaccine containing a subtype 1 strain were examined in yearling ponies. Complement fixing antibody responses were significantly higher against the subtype 1 vaccine strain than against a subtype 2 virus. Complement fixing antibody responses declined rapidly after the second dose of vaccine and had returned to almost pre-vaccination levels eight weeks after the second dose of vaccine. Complement fixing antibody titres to the heterologous subtype 2 strain increased after each successive dose of va...
Geiser DR, Walker RD.The pathophysiology and histopathology of thermal burns in large animals is very similar to that in humans. Burns are classified as first degree, superficial and deep second degree, third degree, and fourth degree, depending upon the depth of thermal injury. Most severe burns will produce a local and a systemic response--both of which must be properly treated to increase the patient's chances for survival. The systemic response is mainly characterized by hypovolemia, fluid and electrolyte loss, protein loss, pulmonary edema, increased caloric requirements, and depressed immune responses. The l...
Asbury AC, Gorman NT, Foster GW.The addition of serum to uterine secretions was shown to opsonize Streptococcus zooepidemicus and significantly enhance bacterial phagocytosis by equine neutrophils. Treatment of serum by heat inactivation at 56 degrees C, EDTA treatment, and C3 consumption reduced phagocytosis and therefore demonstrated that the process was complement-dependent. The amount of C3 present in uterine secretions was measured in a series of 14 mares infected with Streptococcus zooepidemicus . Ten of the 14 mares had detectable amounts of C3; however, the C3 had been cleaved and rendered nonfunctional. The importan...
Reef VB, Dyson SS, Beech J.Three horses with equine lymphosarcoma were examined because of clinical signs including chronic weight loss, respiratory distress, peripheral edema, and chronic colic. Clinicopathologic findings included evidence of an immune-mediated hemolytic anemia. Immune-mediated thrombocytopenia also was diagnosed in 1 of the horses and suspected in another. One horse died in spite of treatment, 1 died 5 hours after surgical removal of a tumor encircling the jejunum, and 1 was euthanatized because of deteriorating condition. Necropsy of each horse revealed extensive neoplastic infiltration of peripheral...
Perryman LE, Wyatt CR.Equine lymphocytes incubated with Con A and isolated on discontinuous BSA density gradients suppressed mixed lymphocyte reactions in a cell dose- and Con A dose-dependent manner. Suppressor lymphocytes were radiosensitive, even after the initial Con A incubation phase was completed. Suppressor activity was consistently demonstrated using peripheral blood mononuclear leukocytes from normal horses, but was absent in thymus cells and variably present in lymph node cells. Suppressor lymphocytes were present in horses with selective IgM deficiency, and within neoplastic lymph nodes from a horse wit...
Bailey M, Lloyd S, Martin SC, Soulsby EJ.Proliferation in vitro of peripheral blood lymphocytes both from horses infected with Strongylus vulgaris and from helminth-free ponies was observed in the presence of extracts of the fourth and fifth stage larvae and adults of S. vulgaris. In addition, S. vulgaris extracts induced transformation in cultures of peripheral blood lymphocytes from sheep and dogs and in mouse spleen cell cultures. Nylon wool non-adherent, T cell enriched fractions of lymphocytes from both mice and horses were stimulated by the S. vulgaris larval mitogen while no proliferation was observed in cultures containing ny...
Metenier L, Grosclaude J, Meriaux JC.The chief application of blood typing in domestic animals is in the verification of parentage. However, the acquisition of good standardized reagents in sufficient quantity remains an obstacle for the development of this work. The production of monoclonal antibodies directed against blood group determinants offers an attractive means of improving both the quality and quantity of serological reagents, and could facilitate the definition of new specificities. Fusions between a mouse myeloma line and splenocytes from mice immunized with horse red cells have resulted in four hybridomas producing a...
Floyd K, Suter PF, Lutz H.Direct staining of the granules of blood eosinophils by anti-immunoglobulin fluorescein isothiocyanate (FITC) conjugates was observed when feline blood smears were tested for presence of feline leukemia virus (FeLV) antigen by immunofluorescent antibody. When blood smears of other species including swine, horses, cattle, dogs, sheep, birds, and human beings were examined, direct staining of eosinophils by FITC conjugates was also detected. This FITC staining was restricted to eosinophils and was not observed in neutrophils, lymphocytes, and platelets. Direct FITC staining of eosinophils does n...
Johnson JA, Prescott JF, Markham RJ.Six foals were inoculated intrabronchially with a suspension of Corynebacterium equi. Six weeks before this challenge, three foals were vaccinated with a C. equi bacterin. Three foals were unvaccinated controls. All foals developed a severe bronchopneumonia in the inoculated lung, indicating that vaccination was not protective. Three foals (two vaccinated, one control) were killed eight to nine days after infection. One control died on day 9 with lesions of disseminated intravascular coagulation. The remaining two foals (one vaccinated, one control) were killed on day 17. C. equi was cultured ...
McBeath DG, Wells PW, Eyre P, Hanna CJ.This paper attempts to relate the practicalities of vaccine development to the ideals which should be aimed for in a new vaccine. The type of immune response induced is dependent upon the nature of the antigen in the vaccine and the site and timing of its presentation to the immune system. In this respect the influence of age, maternal immunity and antigenic competition are discussed. The possible side effects associated with vaccination are defined and vaccines which are currently available for horses are reviewed. These vaccines are mostly for the prevention of respiratory disease. Finally, ...
Weiser G, Kohn C, Vachon A.Immune-mediated hemolytic anemia was diagnosed in two horses on the basis of regenerative anemia, increased erythrocyte fragility in hypotonic saline, autoagglutination, and a positive direct antiglobulin (Coomb's) test. During steroid therapy partial resolution of the anemia was indicated by rising packed cell volume, macrocytosis, and bone marrow erythroid hyperplasia. Using erythrocyte volume distribution histograms (erythrograms), the regenerative response was characterized by analysis of macrocytic and normocytic erythrocyte subpopulations. In both horses, a gradual net increase of about ...
Lány P, Pospísil Z, Zendulková D, Cíhal P, Jahn P.A mild outbreak of acute respiratory infection was reported in racing horses in the fall of 1995. Four studs were investigated for the sources and routes of infection. In five horses from two herds, virus isolates were obtained which, in preliminary typing experiments, were identified as the influenza A/equi 2 virus. The presence of this illness in all the examined herds was confirmed by a rise in specific antibody titres. The affected animals included both older vaccinated horses and young horses not yet vaccinated. Epidemiological studies suggested that the spread of infection occurred in si...
Ramos MGDSN, Campos SDE, Strauch MA, Ott LC, Macieira DB, de Alencar NX, Lessa DAB.Indisputably, the use of antivenoms for the treatment of snakebite envenoming is beneficial for the victims. However, there are few studies addressing the effect of long-term hyperimmunization in inoculated horses. It is known that the injection of snake venoms and adjuvants leads to local and systemic reactions in horses, but little is known about the response of inflammatory proteins. The aim of this study was to evaluate serum proteins and the electrophoretic profile of horses undergoing crotalid venom hyperimmunization. Twenty horses were divided into two groups: an inoculated group, compr...
Goto H, Yamamoto Y, Ohta C, Shirahata T, Higuchi T, Ohishi H.A total of 305 horse sera collected in the Hidaka district of Hokkaido in the years 1988-90 were tested for the presence of hemagglutination-inhibition (HI) antibodies to A/equine/Newmarket/1/77 (H7N7), A/equine/Tokyo/2/71 (H3N8) and A/equine/Kentucky/1/81 (H3N8, Kentucky) strains of equine influenza (EI) virus. Antibodies to the 3 strains were detected in hardly of the 45 sera from 2-years-old horses which were collected before vaccination. Many of the 51 horses, after vaccination with inactivated EI virus, had HI antibodies to the 3 strains in 37 to 88 per cent. However, the number of positi...
Birdwhistell KE, Hurley DJ, Heins B, Peroni JF.Allogeneic solid organ transplantation is currently the only treatment option for end stage organ disease. The shortage of available donor organs has driven efforts to utilize xenogeneic organs for transplantation. In vitro methods for evaluating immune-compatibility are a quick and low cost means of screening novel tissue products prior to more involved, expensive, and invasive live animal studies. Recently, a new analog of the DNA base thymidine, 5-ethynyl-2'-deoxyuridine (EdU), was developed. It may be used in a fast, efficient and specific means of evaluating cell proliferation via flow cy...
Srivastava SK, Barnum DA.Lymphocyte stimulation was observed in whole equine blood in the presence of phytohaemagglutinin and M protein extracted from a typical strain of Streptococcus equi. Blood samples were collected from several healthy horses and horse and pony foals and cultured in vitro with varying concentrations of phytohaemagglutinin and M protein for several days. Phytohaemagglutinin was found to induce lymphocyte stimulation in these animals. Highest mean stimulation indices in horse foals (49.3 +/- 24.4) and pony foals (54.7 +/- 32.0) were observed with 0.625 and 1.25 micrograms/mL phytohaemagglutinin, re...
Tsunoda RH, Serrano-Recalde EC, Arruda RP, Oliveira BMM, Pinto SCC, Celeghini ECC.The aim was to evaluate effects of addition of pentoxifylline to skimmed milk semen extender on uterine inflammatory response. Thirty-six estrous cycles of 15 mares were randomly divided into five groups for artificial insemination (AI): Control: mimicking the AI procedure (n = 7); Extender: deposition of skimmed milk based extender (n = 7); Extender + PTX: skimmed milk based extender plus pentoxifylline (7.18 mM; n = 8); Semen: semen diluted with extender without pentoxifylline (n = 7), and Semen + PTX: semen diluted with extender containing pentoxifylline (n = 7). M...
Hamouzová P, Drábková Z, Stehlíková Š, Řeháková K, Čížek P, Dobešová O, Jahn P, Doubek J.This study determined Tregs and inflammatory cytokines in BALF and peripheral blood (PB) of adult horses with mild and severe asthma and different BALF cytological inflammation profiles. Horses of diverse breeds with asthma (age range: 2-20 years, n = 24) were divided into groups according to the number of points obtained in a standardized clinical scoring system (mild-moderate equine asthma - MEA, severe equine asthma - SEA) and according to the inflammation type based on cytological finding. Plasma levels of IFN-γ, IL-4, IL-10, IL-17 and MMP-9 in the BALF were determined by ELISA. Tregs ...
Chiaradia E, Gaiti A, Terracina L, Avellini L.Physical exercise induces a reduction of immune defences and an imbalance of red-ox status. In this study plasma levels of cysteine and homocysteine (Hcy) were determined in horses before and after submaximal treadmill exercise as well as the effect on horse lymphocyte proliferation. The exercise induced a significant increase in plasma Hcy levels, which remained high both after the 20 min recovery period and after 2 h of rest. Moreover, a reduction of lymphocyte responsiveness to the proliferative stimulus induced by Concanavalin A was observed. The effects of different Hcy concentrations on ...
Marr KA, Fairbairn SM, Page CP, Lees P, Cunningham FM.Antigen challenge involving exposure to straw and mouldy hay for 7 h produced lung function changes and neutrophil recruitment to the lungs in horses with chronic obstructive pulmonary disease (COPD). During the challenge, an increase in radiolabelled neutrophils in the lungs occurred, together with increased respiratory rate and pleural pressure. The role of platelet activating factor (PAF) in antigen-induced neutrophil accumulation, and increased pleural pressure and respiratory rate was investigated by administering the PAF receptor antagonist WEB 2086 to asymptomatic COPD horses prior to a...
Watson ED, Sertich PL, Zanecosky HG.Ovulation has been likened to an inflammatory process. Inflammatory cells accumulate in the ovulating follicle, presumably because of chemotactic factors. Chemotactic activity was measured in fluid aspirated from follicles of estrous mares 0, 12, 24, and 36 hours after ultrasonographic detection of a 35-mm follicle and IV treatment with 2,500 IU of human chorionic gonadotropin. Chemotaxis was assessed by measuring directional migration of equine neutrophils under agarose. Follicular fluid acted as a chemoattractant for neutrophils, but there was no significant difference in chemotactic activit...
du Toit N, McGorum BC, Pemberton AD, Brown J, Dacre KJ.Cyathostomins (Cyathostominae) are regarded as the most pathogenic equine nematode worldwide. These nematodes are difficult to control in equine populations due to emerging anthelmintic resistance and evasion of encysted larval cyathostomins to regular modern anthelmintics. Mast cells and their proteinases have been shown to play a role in the mammalian immune response to nematode infections. Involvement of mast cells and mast cell proteinases in the equine immune response to cyathostomin infection is proposed. A technique was established to perform immunohistochemical staining using polyclona...
Watson ED.The ability of equine endometrium to release prostaglandin (PG) F, PGE2, and leukotriene (LT) B4 was studied in vitro, using endometrial tissue from diestrous mares. Because of the high cross-reactivity of the PGF antiserum with PGF1 alpha and with PGF2 alpha, results were quoted as total immunoreactive PGF. Significant concentrations of these arachidonate metabolites were released into tissue culture medium between 1 and 24 hours of incubation. Significantly higher concentrations of PGE, but not of PGE2 or LTB4, were released from endometria of mares with chronic endometritis than from genita...
Hamouzová P, Dobešová O, Řeháková K, Stehlíková Š, Čížek P, Drábková Z, Jahn P, Doubek J.This study is the first to provide information on the lymphocyte subpopulations in peritoneal effusions in horses. Peritoneal transudates (n = 12), peritoneal exudates (n = 6) and a pleural exudate (n = 1) were analyzed. The total nucleated cell count (TNCC), total protein (TP) and matrix metalloproteinase-9 (MMP-9) concentration determined by ELISA were measured and routine cytological evaluation was performed. CD3, CD4, CD8 and CD21 positive cells were detected by flow cytometry. A higher percentage of neutrophils (P < 0.05) and higher MMP-9 (P < 0.01) levels were found in ex...
BonenClark GD, MacKay RJ, Ward RE, Sheerin B.To evaluate the humoral response of horses to vaccination, using a murine monoclonal anti-idiotypic antibody (A4) that shares an epitope with lipid A. Methods: Serum concentrations of antilipid A antibody and 1C9 (epitope on murine monoclonal antilipid A antibody) were measured serially during the period of vaccination with A4. Methods: 6 clinically normal adult ponies. Methods: Ponies were inoculated IM 3 times at monthly intervals with A4. Two weeks after each inoculation, serum was obtained and was assayed by ELISA for antilipid A and 1C9 concentrations. Additional vaccinations were given t...
Mancini DA, Pereira AS, Mendonça RM, Kawamoto AH, Alves RC, Pinto JR, Mori E, Richtzenhain LJ, Mancini-Filho J.Equines are susceptible to respiratory viruses such as influenza and parainfluenza. Respiratory diseases have adversely impacted economies all over the world. This study was intended to determine the presence of influenza and parainfluenza viruses in unvaccinated horses from some regions of the state of São Paulo, Brazil. Blood serum collected from 72 equines of different towns in this state was tested by hemagglutination inhibition test to detect antibodies for both viruses using the corresponding antigens. About 98.6% (71) and 97.2% (70) of the equines responded with antibody protective tit...
Cunningham FM, Vandergrifft E, Bailey SR, Sepulveda MF, Goode NT, Horohov DW.The cytokine, interleukin (IL)-5 stimulates eosinophil differentiation, activation and survival and can prime these cells, increasing the response to other mediators. In view of its many effects on eosinophils, IL-5 has been implicated in the pathogenesis of allergic disease in man. Here we report the cloning of equine IL-5 and expression of the recombinant protein by transfection of Chinese hamster ovary (CHO) cells. The cloned cDNA sequence consisted of 405 nucleotides and encoded a protein of 135 amino acids. There is >85% identity with feline, bovine, ovine, canine, and human IL-5 seque...
Holmes RA, Klei TR, McClure JR, Turk MA, Watters JW, Chapman MR.Semiselective mesenteric arteriography was performed at regular intervals (inoculation weeks [IW] 0, 11, 18, and 24) in 9 of 10 pony foals raised to be free of parasites. Fifty infective larvae (L3) of Strongylus vulgaris were administered weekly for 4 weeks, then every 2 weeks through the 20th week. Three ponies were given ivermectin (oral paste, 0.2 mg/kg of body weight) treatment at IW 8, 16 and 24. Four ponies were inoculated, but did not receive ivermectin, and a third group of 2 ponies acted as uninoculated controls. Control ponies did not have gross or arteriographic lesions, whereas th...
Williams MR.Rats were injected intradermally with rabbit anti-ovalbumin serum and 3 hours later were challenged intravenously with ovalbumin and Evans Blue dye. Inflammatory lesions were produced within 20 minutes and their size was markedly dose-dependent. Attempts were made to interfere with this passive cutaneous anaphylaxis (PCA) by admixture of normal IgG with the rabbit anti-ovalbumin to measure the relative tissue binding affinities of IgG from various species. It was found that normal IgG from any of the species tested had an enhancing effect on PCA in rats. These immunoglobulins serially arranged...
Ahmed JS, Schmid G, Hörchner F.Pony peripheral blood lymphocytes (PBL) were stimulated with a soluble fraction of Trypanosoma (T.) evansi (SF). As determined by 3H-thymidine incorporation, the cells underwent a proliferative response and were able to: a) produce a factor having the biological activities of interleukin 2 (IL-2) since their supernatants could support the in vitro growth of pony PBL stimulated with concanavalin A (Con A-blasts); b) undergo a further proliferative response when incubated in short term cultures with SF, human recombinant IL-2 (hrIL-2), or both c) bind specifically radiolabelled hrIL-2 (125I-hrIL...
Meng Q, Lin Y, Ma J, Ma Y, Zhao L, Li S, Yang K, Zhou J, Shen R, Zhang X, Shao Y.Data from successful attenuated lentiviral vaccine studies indicate that fully mature Env-specific antibodies characterized by high titer, high avidity, and the predominant recognition of conformational epitopes are associated with protective efficacy. Although vaccination with a DNA prime/recombinant vaccinia-vectored vaccine boost strategy has been found to be effective in some trials with non-human primate/simian/human immunodeficiency virus (SHIV) models, it remains unclear whether this vaccination strategy could elicit mature equine infectious anemia virus (EIAV) Env-specific antibodies, ...
Camp CJ, Leid HW.Extracts of Onchocerca cervicalis, an equine parasite, were incubated with radiolabeled equine neutrophils and neutrophil migration was assessed for factors derived from the parasite itself or for host-derived factors after incubation of these same parasite extracts with equine serum. No stimulus for cell migration was observed in saline extracts of adult worms, uterine microfilariae, or skin microfilariae at any dosage tested. However, after incubation of saline extracts with fresh normal equine sera a marked stimulus for neutrophil migration was observed. Ablation of this biologic activity w...
Kalpokas I, Martínez MN, Cavestany D, Perdigón F, Mattos RC, Meikle A.We investigated the early effects of the equine embryo on maternal serum concentrations of insulin-like growth factor 1 (IGF1), leptin and adiponectin, uterine immune cells and genes and proteins related to embryo development and the maintenance of pregnancy. Ipsilateral endometrial expression was assessed on Days 7 and 13 after ovulation for the following transcripts: oestrogen receptor ERα (ESR1), progesterone receptor (PGR), progestin and adipoQ receptor family member 5 (PAQR5), oxytocin receptor (OXTR), prostaglandin-endoperoxide synthase 2 (PTGS2), raf-1 proto-oncogene serine/threonine k...
Page AE, Adam E, Stewart JC, Gonzales C, Barker V, Horohov DW.While the use of lipopolysaccharide (LPS) to induce inflammation has been well described in the horse, the object of this study was to evaluate the effect of repeated intra-articular LPS injections and determine whether this method may be of use to assess changes in gene expression related to inflammation. Six mixed breed horses were utilized for this study, with three horses aged 10-17 years (older group) and three horses aged 3 years (younger group). One milliliter of phosphate-buffered saline containing 3 μg of LPS from Escherichia coli O111:B4 was aseptically injected into either the radi...
Palmer JE, Benson CE, Lotz GW.Ten healthy, mature ponies were orally infected with Ehrlichia risticii using Ehrlichia-infected P388D1 mouse monocyte tissue culture cells. Seven developed signs of equine ehrlichial colitis including fever, depression, anorexia, reduced borborygmi, increased abdominal hyperresonance, and diarrhoea. Three remained clinically normal apart from early fever in one. Indirect fluorescent antibody titres were detected in the clinically affected ponies by Days 12 to 17 post infection and increased rapidly to high levels (1:640 to 1:5120) which were maintained until the end of the observation period ...
Troedsson MH, Lee CS, Franklin RD, Crabo BG.The effect of seminal plasma on polymorphonuclear neutrophil (PMN) chemotaxis, PMN phagocytosis and complement-induced cytolysis was determined using blood plasma pooled from four horses and seminal plasma pooled from two stallions. To investigate chemotaxis, complement in blood plasma was activated with E. coli lipopolysaccharide in the presence of 0-50% seminal plasma diluted with a standardized volume of McCoy's medium and placed in a chemotactic chamber. Chemotaxis of blood derived equine PMNs toward the chemoattractants was determined after incubation at 37 degrees C for 45 min. To invest...
Escribano BM, Agüera EI, Vivo R, Santisteban R, Castejón FM, Rubio MD.The aim of this work was to assess whether progressive training caused an improvement in the nonspecific immune response of colts because several unusual infections are due to defects inherent in the neutrophilic function among which respiratory diseases are a major defect in the performance of athletes taking part in professional sports activities. A group of 7 Anglo-Arabian colts belonging to the Army was selected. These animals carry out training programmes for their participation in National Jumping Competitions. During a submaximal exercise test (heart rate 150 beats/min and lactate level...
Han X, Quan Y, Gao X, Xiang W, Zhou J.To elucidate the role of N-glycosylation in fetal donkey dermal cell (FDD)-attenuated equine infectious anemia virus (EIAV), we constructed an N-glycosylation reverse-mutation molecular clone, pLGN191N236N246. This viral molecular clone was derived from the infectious clone pLGFD3-8 by site-directed mutagenesis. This clone was used to transfect fetal donkey dermal (FDD) cells. Infectious characteristics of transfectants were monitored by RT-PCR, indirect immune fluorescence and reverse transcriptase activity assay. After three passages in FDD cells, viral replications in the supernatant of cel...
Sanada Y, Noda H, Nagahata H.The blastogenic response of lymphocytes from 16 newborn foals naturally infected with Rhodococcus equi was investigated, in order to evaluate the relationship between R. equi infection and depressed host response. Naturally infected foals showed evidence of R. equi infection at 5-6 weeks of age, as determined by clinical, haematological, bacteriological and serological methods. The blastogenic response of lymphocytes against phytohaemagglutinin was significantly depressed (stimulation index < 1.80; P < 0.01, P < 0.05) in R. equi-infected foals at 5-6 weeks of age compared with those o...
Valiev AG, Valieva TA, Valeeva GR, Speranskiĭ VV, Levachev MM.The influence of essential fatty acids of mare's milk fat on the immunocompetent system and nonspecific resistance of male rats weighing 135-145 g was investigated after 6 weeks of feeding. Rats were fed with isocaloric purified diets containing 15% of test fat (in control--combination of lard and sunflower oil) which provide the ratio of omega-6/omega-3 fatty acids equal to 0.76. An increase of immune responsiveness and nonspecific resistance in the group fed with the diet with mare's milk fat on day 6 after a single immunization of the animals with 5% sheep erythrocyte suspension was noted.
Barbey C, Cauchard S, Cauchard J, Laugier C, Hartke A, Petry S.Rhodococcus equi remains a significant pathogen, causing severe pneumonia in foals. The development of vaccines and serologic diagnosis could be greatly facilitated by studying the humoral immune response to this equine pathogen. In this study, a crude extract of R. equi ATCC 33701-secreted proteins combined with the Montanide® ISA70 adjuvant was found to be highly immunogenic in mice with the highest titer of 99,000 on day 42 after the first subcutaneous immunization. This immune response was dependent on the quantity of proteins injected and the presence of adjuvant. By dot-blotting, eight ...
