Understanding stress concentration about a nutrient foramen.
Journal of biomechanics    September 23, 2003   Volume 36, Issue 10 1511-1521 doi: 10.1016/s0021-9290(03)00130-1
Götzen N, Cross AR, Ifju PG, Rapoff AJ.We investigated the microstructural basis of a reduced stress concentration around the primary nutrient foramen of the equine third metacarpus. We quantified the spatial variations of compositional parameters (mineral content, volume fraction, histological architecture, and osteonal trajectories) from microradiographs and polarizing microscopic images of thin sections. These variations in composition and organization in turn cause variations in mechanical properties of cortical bone. We modeled the spatially inhomogeneous anisotropic elastic properties based on the measured compositional param...

The Journal of general virology    September 19, 2003   Volume 84, Issue Pt 10 2625-2634 doi: 10.1099/vir.0.19268-0
Soboll G, Whalley JM, Koen MT, Allen GP, Fraser DG, Macklin MD, Swain WF, Lunn DP.Equine herpesvirus-1 (EHV-1) causes serious disease in horses throughout the world, despite the frequent use of vaccines. CTLs are thought to be critical for protection from primary and reactivating latent EHV-1 infections. However, the antigen-specificity of EHV-1-specific CTLs is unknown. The aim of this study was to identify EHV-1 genes that encode proteins containing CTL epitopes and to determine their MHC I (or ELA-A in the horse) restriction. Equine dendritic cells, transfected with a series of EHV-1 genes, were used to stimulate autologous CTL precursor populations derived from previous...

Research in veterinary science    September 18, 2003   Volume 75, Issue 3 223-229 doi: 10.1016/s0034-5288(03)00116-4
Dowdall SM, Proudman CJ, Love S, Klei TR, Matthews JB.Cyathostomins are important equine gastrointestinal parasites. Mass emergence of mucosal stage larvae causes a potentially fatal colitis. Mucosal stages are undetectable non-invasively. An assay that would estimate mucosal larval stage infection would greatly assist in treatment, control and prognosis. Previously, we identified two putative diagnostic antigens (20 and 25 kDa) in somatic larval preparations. Here, we describe their purification and antigen-specific IgG(T) responses to them. Western blots confirmed the purity of the antigens and showed that epitopes in the 20 kDa complex were sp...

Journal of animal science    September 13, 2003   Volume 81, Issue 9 2311-2321 doi: 10.2527/2003.8192311x
Cartmill JA, Thompson DL, Storer WA, Gentry LR, Huff NK.Previous observations from this laboratory indicated that horses with high BCS could have resting plasma leptin concentrations ranging from low (1 to 5 ng/mL) to very high (10 to 50 ng/mL). To study the possible interactions of leptin secretion with other endocrine systems, BCS and plasma leptin concentrations were measured on 36 mares and 18 geldings. From mares and geldings that had a mean BCS of at least 7.5, five with the lowest (low leptin) and five with the highest (high leptin) leptin concentrations were selected. Jugular blood samples were collected twice daily for 3 d from the 20 sele...
Thyrotropin releasing hormone interactions with growth hormone secretion in horses.
Journal of animal science    September 13, 2003   Volume 81, Issue 9 2343-2351 doi: 10.2527/2003.8192343x
Pruett HE, Thompson DL, Cartmill JA, Williams CC, Gentry LR.Light horse mares, stallions, and geldings were used to 1) extend our observations on the thyrotropin releasing hormone (TRH) inhibition of GH secretion in response to physiologic stimuli and 2) test the hypothesis that stimulation of endogenous TRH would decrease the normal rate of GH secretion. In Exp. 1 and 2, pretreatment of mares with TRH (10 microg/kg BW) decreased (P < 0.001) the GH response to exercise and aspartate infusion. Time analysis in Exp. 3 indicated that the TRH inhibition lasted at least 60 min but was absent by 120 min. Administration of a single injection of TRH to stal...

Journal of animal science    September 13, 2003   Volume 81, Issue 9 2285-2293 doi: 10.2527/2003.8192285x
Pfister JA, Stegelmeier BL, Gardner DR, James LF.Spotted locoweed (Astragalus lentiginosus var. diphysus) is a toxic, perennial plant that may, if sufficient precipitation occurs, dominate the herbaceous vegetation of pinyon-juniper woodlands on the Colorado Plateau. Six cow/calf pairs and four horses grazed a 20-ha pasture with dense patches of locoweed in eastern Arizona during spring 1998. Locoweed density was 0.7 plants/m2 in the pasture. Locoweed averaged 30.4% NDF and 18.4% CP. Concentrations of the locoweed toxin, swainsonine, fluctuated from 1.25 to 2 mg/g in locoweed. Horses ate more (P < 0.01) bites of locoweed than did cows (15...
Characterization of RNA elements that regulate gag-pol ribosomal frameshifting in equine infectious anemia virus.
Journal of virology    September 13, 2003   Volume 77, Issue 19 10280-10287 doi: 10.1128/jvi.77.19.10280-10287.2003
Chen C, Montelaro RC.Synthesis of Gag-Pol polyproteins of retroviruses requires ribosomes to shift translational reading frame once or twice in a -1 direction to read through the stop codon in the gag reading frame. It is generally believed that a slippery sequence and a downstream RNA structure are required for the programmed -1 ribosomal frameshifting. However, the mechanism regulating the Gag-Pol frameshifting remains poorly understood. In this report, we have defined specific mRNA elements required for sufficient ribosomal frameshifting in equine anemia infectious virus (EIAV) by using full-length provirus rep...

In expt 1, three groups of five mares were tested: intact-control, intact-treated and ovariectomized-treated mares. In expt 2, mares previously subjected to artificial pho...
Quantitative analysis of eumelanin and pheomelanin in humans, mice, and other animals: a comparative review.
Pigment cell research    September 3, 2003   Volume 16, Issue 5 523-531 doi: 10.1034/j.1600-0749.2003.00072.x
Ito S, Wakamatsu K.The color of hair, skin, and eyes in animals mainly depends on the quantity, quality, and distribution of the pigment melanin, which occurs in two types: black to brown eumelanin and yellow to reddish pheomelanin. Microanalytical methods to quantify the amounts of eumelanin and pheomelanin in biological materials were developed in 1985. The methods are based on the chemical degradation of eumelanin to pyrrole-2,3,5-tricarboxylic acid and of pheomelanin to aminohydroxyphenylalanine isomers, which can be analyzed and quantitated by high performance liquid chromatography. This review summarizes a...

Virus research    September 3, 2003   Volume 96, Issue 1-2 141-145 doi: 10.1016/s0168-1702(03)00175-8
Chambers G, Ellsmore VA, O'Brien PM, Reid SW, Love S, Campo MS, Nasir L.The equine sarcoid, one of the most common dermatological lesions in equids, is a benign, locally invasive dermal fibroblastic lesion. Previous studies have suggested an association with two bovine papilloma virus (BPV) types, BPV-1 and BPV-2. In the present study, we examined sarcoids from horses from two geographical areas, Switzerland and the UK, for the major transforming gene of BPV, E5. We detected BPV DNA for the E5 open reading frame and viral E5 RNA transcripts in most sarcoids. Sequence analysis of the E5 open reading frame of sarcoid-associated BPV detected several unique DNA sequen...
