Development of multiplex gold nanoparticles biosensors for ultrasensitive detection and genotyping of equine herpes viruses.
Scientific reports    September 13, 2023   Volume 13, Issue 1 15140 doi: 10.1038/s41598-023-41918-4
Ghoniem SM, ElZorkany HE, Hagag NM, El-Deeb AH, Shahein MA, Hussein HA.Gold nanoparticles (GNPs) biosensors can detect low viral loads and differentiate between viruses types, enabling early diagnosis and effective disease management. In the present study, we developed GNPs biosensors with two different capping agent, citrate-GNPs biosensors and polyvinylpyrrolidone (PVP)-GNPs biosensors for detection of EHV-1 and EHV-4 in multiplex real time PCR (rPCR). Citrate-GNPs and PVP-GNPs biosensors can detect dilution 10 of EHV-1 with mean Cycle threshold (Ct) 11.7 and 9.6, respectively and one copy as limit of detection, while citrate-GNPs and PVP-GNPs biosensors can de...
Clostridioides difficile infection in thoroughbred horses in Japan from 2010 to 2021.
Scientific reports    August 11, 2023   Volume 13, Issue 1 13099 doi: 10.1038/s41598-023-40157-x
Uchida-Fujii E, Niwa H, Senoh M, Kato H, Kinoshita Y, Mita H, Ueno T.We encountered 34 Clostridioides difficile (C. difficile) infection (CDI) cases among Thoroughbred horses in Japan from 2010 to 2021. Among them, 79.4% (27/34) either died or were euthanised. The risk factors associated with CDI and mortality among Japanese Thoroughbred horses remain unclear. We used genetic methods to examine C. difficile strains and their relationships with prognosis. Twenty-two (64.7%) cases were hospitalised at the onset of colitis. Outcomes were balanced for hospitalisation rates at the onset of colitis. The mortality rates of cases treated with metronidazole (65.0%) were...

Scientific reports    June 27, 2023   Volume 13, Issue 1 10385 doi: 10.1038/s41598-023-37295-7
Krishnan BS, Jones LR, Elmore JA, Samiappan S, Evans KO, Pfeiffer MB, Blackwell BF, Iglay RB.Visible and thermal images acquired from drones (unoccupied aircraft systems) have substantially improved animal monitoring. Combining complementary information from both image types provides a powerful approach for automating detection and classification of multiple animal species to augment drone surveys. We compared eight image fusion methods using thermal and visible drone images combined with two supervised deep learning models, to evaluate the detection and classification of white-tailed deer (Odocoileus virginianus), domestic cow (Bos taurus), and domestic horse (Equus caballus). We cla...
Fatty acid fingerprints in bronchoalveolar lavage fluid and its extracellular vesicles reflect equine asthma severity.
Scientific reports    June 17, 2023   Volume 13, Issue 1 9821 doi: 10.1038/s41598-023-36697-x
Höglund N, Nieminen P, Mustonen AM, Käkelä R, Tollis S, Koho N, Holopainen M, Ruhanen H, Mykkänen A.Equine asthma (EA) is an inflammatory disease of the lower airways driven by mediators released from cells. Extracellular vesicles (EVs) are vehicles for lipid mediators, which possess either pro-inflammatory or dual anti-inflammatory and pro-resolving functions. In this study, we investigated how the respiratory fatty acid (FA) profile reflects airway inflammatory status. The FA composition of bronchoalveolar lavage fluid (BALF), BALF supernatant, and bronchoalveolar EVs of healthy horses (n = 15) and horses with mild/moderate EA (n = 10) or severe EA (SEA, n = 5) was determined w...

Scientific reports    June 17, 2023   Volume 13, Issue 1 9858 doi: 10.1038/s41598-023-37069-1
This study aimed to evaluate the change of white blood cell count, serum concentration of cortisol, C-reactive protein, albumin and globulin fractions in horse after road transport, and to assess the linkage among hypothalamic-pituitary-adrenal axis (HPA) and inflammatory reaction. From 10 horses blood samples were collected at rest, before road transport (218 km) (BT); after unloaded (AT), 30 and 60 min after unloaded (AT30 and AT60) in order to assess white blood cell count (WBC), serum cortisol, C-reactive protein (CRP), total proteins, albumin, α1-, α-2, β1-, β2- and γ-globulins. WB...
Refining the evolutionary tree of the horse Y chromosome.
Scientific reports    June 2, 2023   Volume 13, Issue 1 8954 doi: 10.1038/s41598-023-35539-0
Bozlak E, Radovic L, Remer V, Rigler D, Allen L, Brem G, Stalder G, Castaneda C, Cothran G, Raudsepp T, Okuda Y, Moe KK, Moe HH, Kounnavongsa B....The Y chromosome carries information about the demography of paternal lineages, and thus, can prove invaluable for retracing both the evolutionary trajectory of wild animals and the breeding history of domesticates. In horses, the Y chromosome shows a limited, but highly informative, sequence diversity, supporting the increasing breeding influence of Oriental lineages during the last 1500 years. Here, we augment the primary horse Y-phylogeny, which is currently mainly based on modern horse breeds of economic interest, with haplotypes (HT) segregating in remote horse populations around the wor...
New genomic insights into the conformation of Lipizzan horses.
Scientific reports    June 2, 2023   Volume 13, Issue 1 8990 doi: 10.1038/s41598-023-36272-4
Gmel AI, Brem G, Neuditschko M.Conformation traits are important selection criteria in equine breeding, as they describe the exterior aspects of the horse (height, joint angles, shape). However, the genetic architecture of conformation is not well understood, as data of these traits mainly consist of subjective evaluation scores. Here, we performed genome-wide association studies on two-dimensional shape data of Lipizzan horses. Based on this data, we identified significant quantitative trait loci (QTL) associated with cresty neck on equine chromosome (ECA)16 within the MAGI1 gene, and with type, hereby differentiating heav...
Horses (Equus caballus) facial micro-expressions: insight into discreet social information.
Scientific reports    May 27, 2023   Volume 13, Issue 1 8625 doi: 10.1038/s41598-023-35807-z
Tomberg C, Petagna M, de Selliers de Moranville LA.Facial micro-expressions are facial expressions expressed briefly (less than 500 ms) and involuntarily. Described only in humans, we investigated whether micro-expressions could also be expressed by non-human animal species. Using the Equine Facial action coding system (EquiFACS), an objective tool based on facial muscles actions, we demonstrated that a non-human species, Equus caballus, is expressing facial micro-expressions in a social context. The AU17, AD38 and AD1 were selectively modulated as micro-expression-but not as standard facial expression (all durations included)-in presence of ...
Circulating miR-146b and miR-27b are efficient biomarkers for early diagnosis of Equidae osteoarthritis.
Scientific reports    May 17, 2023   Volume 13, Issue 1 7966 doi: 10.1038/s41598-023-35207-3
Yassin AM, AbuBakr HO, Abdelgalil AI, Farid OA, El-Behairy AM, Gouda EM.One of the most orthopedic problems seen in the equine is osteoarthritis (OA). The present study tracks some biochemical, epigenetic, and transcriptomic factors along different stages of monoiodoacetate (MIA) induced OA in donkeys in serum and synovial fluid. The aim of the study was the detection of sensitive noninvasive early biomarkers. OA was induced by a single intra-articular injection of 25 mg of MIA into the left radiocarpal joint of nine donkeys. Serum and synovial samples were taken at zero-day and different intervals for assessment of total GAGs and CS levels as well as miR-146b, m...
Water stabilizes an alternate turn conformation in horse heart myoglobin.
Scientific reports    April 13, 2023   Volume 13, Issue 1 6094 doi: 10.1038/s41598-023-32821-z
Bronstein A, Marx A.Comparison of myoglobin structures reveals that protein isolated from horse heart consistently adopts an alternate turn conformation in comparison to its homologues. Analysis of hundreds of high-resolution structures discounts crystallization conditions or the surrounding amino acid protein environment as explaining this difference, that is also not captured by the AlphaFold prediction. Rather, a water molecule is identified as stabilizing the conformation in the horse heart structure, which immediately reverts to the whale conformation in molecular dynamics simulations excluding that structur...
Equine odontoclastic tooth resorption and hypercementosis (EOTRH): microspatial distribution of trace elements in hypercementosis-affected and unaffected hard dental tissues.
Scientific reports    March 28, 2023   Volume 13, Issue 1 5048 doi: 10.1038/s41598-023-32016-6
Wright AL, Earley ET, Austin C, Arora M.Equine Odontoclastic Tooth Resorption and Hypercementosis (EOTRH) is a common, painful and poorly understood disease. Enamel, dentin and cementum accumulate both essential and toxic trace elements during mineralization. Characterization of the spatial accumulation pattern of trace elements may provide insight into the role that toxic elements play and inform biological processes affecting these hard dental tissues for future research. Laser ablation-inductively coupled plasma-mass spectrometry (LA-ICP-MS) was used to map the distribution of multiple trace elements and heavy metals across equin...

Scientific reports    March 21, 2023   Volume 13, Issue 1 4616 doi: 10.1038/s41598-023-31540-9
Diel de Amorim M, Dong L, Byron M, Foster RA, Klein C, Saleh M, Saleh T, Card C.Oxytocin is a hormone with functions in: reproduction, maternal bonding, milk ejection, and feeding/social behavior, and is reported to be present in a variety of tissues. Our goal is to characterize oxytocin and leucyl and cystinyl aminopeptidase (LNPEP/oxytocinase), a key regulator of oxytocin in mares. We measured serum and tissue LNPEP by ELISA from ovulation (D0) until D21-22 in non-pregnant (n = 5) and pregnant mares (n = 6); and in periparturient and postpartum mares (n = 18). Placenta (n = 7) and homogenized tissue of diestrus mares (n = 6) were evaluated using prot...
Risk factors for, and prediction of, exertional heat illness in Thoroughbred racehorses at British racecourses.
Scientific reports    March 14, 2023   Volume 13, Issue 1 3063 doi: 10.1038/s41598-023-27892-x
Trigg LE, Lyons S, Mullan S.The development of exertional heat illness (EHI) is a health, welfare and performance concern for racehorses. However, there has been limited multivariable assessment of the possible risk factors for EHI in racehorses, despite such information being vital for regulators to effectively manage the condition. Consequently, this study aimed to identify the risk factors associated with the occurrence of EHI in Thoroughbred racehorses and assess the ability of the risk factor model to predict the occurrence of EHI in racehorses to assist in early identification. Runners at British racecourses record...

Scientific reports    March 10, 2023   Volume 13, Issue 1 4029 doi: 10.1038/s41598-023-31173-y
Langreder N, Schäckermann D, Meier D, Becker M, Schubert M, Dübel S, Reinard T, Figge-Wegener S, Roßbach K, Bäumer W, Ladel S, Hust M.Insect bite hypersensitivity (IBH) is the most common allergic skin disease of horses. It is caused by insect bites of the Culicoides spp. which mediate a type I/IVb allergy with strong involvement of eosinophil cells. No specific treatment option is available so far. One concept could be the use of a therapeutic antibody targeting equine interleukin 5, the main activator and regulator of eosinophils. Therefore, antibodies were selected by phage display using the naïve human antibody gene libraries HAL9/10, tested in a cellular in vitro inhibition assay and subjected to an in vitro affinity...
Horses cross-modally recognize women and men.
Scientific reports    March 8, 2023   Volume 13, Issue 1 3864 doi: 10.1038/s41598-023-30830-6
Gouyet C, Ringhofer M, Yamamoto S, Jardat P, Parias C, Reigner F, Calandreau L, Lansade L.Several studies have shown that horses have the ability to cross-modally recognize humans by associating their voice with their physical appearance. However, it remains unclear whether horses are able to differentiate humans according to different criteria, such as the fact that they are women or men. Horses might recognize some human characteristics, such as sex, and use these characteristics to classify them into different categories. The aim of this study was to explore whether domesticated horses are able to cross-modally recognize women and men according to visual and auditory cues, using...

Scientific reports    March 6, 2023   Volume 13, Issue 1 3703 doi: 10.1038/s41598-023-30875-7
Fisher M, Nebroski M, Davies J, Janzen E, Sullivan D, Lung O.The complete genome of a novel torque teno virus species (Torque teno equus virus 2 (TTEqV2) isolate Alberta/2018) was obtained by high-throughput sequencing (HTS) of nucleic acid extracted from the lung and liver tissue of a Quarter Horse gelding that died of nonsuppurative encephalitis in Alberta, Canada. The 2805 nucleotide circular genome is the first complete genome from the Mutorquevirus genus and has been approved as a new species by the International Committee on Taxonomy of Viruses. The genome contains several characteristic features of torque teno virus (TTV) genomes, including an OR...
Horses discriminate human body odors between fear and joy contexts in a habituation-discrimination protocol.
Scientific reports    February 25, 2023   Volume 13, Issue 1 3285 doi: 10.1038/s41598-023-30119-8
Jardat P, Destrez A, Damon F, Menard-Peroy Z, Parias C, Barrière P, Keller M, Calandreau L, Lansade L.Animals are widely believed to sense human emotions through smell. Chemoreception is the most primitive and ubiquitous sense, and brain regions responsible for processing smells are among the oldest structures in mammalian evolution. Thus, chemosignals might be involved in interspecies communication. The communication of emotions is essential for social interactions, but very few studies have clearly shown that animals can sense human emotions through smell. We used a habituation-discrimination protocol to test whether horses can discriminate between human odors produced while feeling fear vs....
Training drives turnover rates in racehorse proximal sesamoid bones.
Scientific reports    January 27, 2023   Volume 13, Issue 1 205 doi: 10.1038/s41598-022-26027-y
Shaffer SK, Stover SM, Fyhrie DP.Focal bone lesions are often found prior to clinically relevant stress-fractures. Lesions are characterized by low bone volume fraction, low mineral density, and high levels of microdamage and are hypothesized to develop when bone tissue cannot sufficiently respond to damaging loading. It is difficult to determine how exercise drives the formation of these lesions because bone responds to mechanical loading and repairs damage. In this study, we derive steady-state rate constants for a compartment model of bone turnover using morphometric data from fractured and non-fractured racehorse proximal...
TLR4 and MD2 variation among horses with differential TNFα baseline concentrations and response to intravenous lipopolysaccharide infusion.
Scientific reports    January 27, 2023   Volume 13, Issue 1 1486 doi: 10.1038/s41598-023-27956-y
Mukhopadhyay A, Cook SR, SanMiguel P, Ekenstedt KJ, Taylor SD.Gram-negative bacterial septicemia is mediated through binding of lipopolysaccharide (LPS) to mammalian toll-like receptor protein 4 (TLR4). TLR4 and its cognate protein, myeloid differentiation factor 2 (MD2) form a heterodimeric complex after binding LPS. This complex induces a cascade of reactions that results in increased proinflammatory cytokine gene expression, including TNFα, which leads to activation of innate immunity. In horses, the immune response to LPS varies widely. To determine if this variation is due to differences in TLR4 or MD2, DNA from 15 healthy adult horses with differe...
Prediction of continuous and discrete kinetic parameters in horses from inertial measurement units data using recurrent artificial neural networks.
Scientific reports    January 13, 2023   Volume 13, Issue 1 740 doi: 10.1038/s41598-023-27899-4
Parmentier JIM, Bosch S, van der Zwaag BJ, Weishaupt MA, Gmel AI, Havinga PJM, van Weeren PR, Braganca FMS.Vertical ground reaction force (GRFz) measurements are the best tool for assessing horses' weight-bearing lameness. However, collection of these data is often impractical for clinical use. This study evaluates GRFz predicted using data from body-mounted IMUs and long short-term memory recurrent neural networks (LSTM-RNN). Twenty-four clinically sound horses, equipped with IMUs on the upper-body (UB) and each limb, walked and trotted on a GRFz measuring treadmill (TiF). Both systems were time-synchronised. Data from randomly selected 16, 4, and 4 horses formed training, validation, and test dat...

Scientific reports    January 6, 2023   Volume 13, Issue 1 300 doi: 10.1038/s41598-023-27374-0
Muko R, Sunouchi T, Urayama S, Toishi Y, Kusano K, Sato H, Muranaka M, Shin T, Oikawa MA, Ojima Y, Ali M, Nomura Y, Matsuda H, Tanaka A.Histidine-rich glycoprotein (HRG) is abundant plasma protein with various effects on angiogenesis, coagulation, and immune responses. Previously, we identified the base and amino acid sequences of equine HRG (eHRG) and revealed that eHRG regulates neutrophil functions. In this study, we first conducted a large-scale gene analysis with DNA samples extracted from 1700 Thoroughbred horses and identified unique insertion/deletion polymorphisms in the histidine-rich region (HRR) of eHRG. Here we report two types of polymorphisms (deletion type 1 [D1] and deletion type 2 [D2]) containing either a 45...
DIA mass spectrometry characterizes urinary proteomics in neonatal and adult donkeys.
Scientific reports    December 30, 2022   Volume 12, Issue 1 22590 doi: 10.1038/s41598-022-27245-0
Yu F, Chen Y, Liu B, Wang T, Ding Z, Yi Z, Zhu Y, Li J.Health monitoring is critical for newborn animals due to their vulnerability to diseases. Urine can be not only a useful and non-invasive tool (free-catch samples) to reflect the physiological status of animals but also to help monitor the progression of diseases. Proteomics involves the study of the whole complement of proteins and peptides, including structure, quantities, functions, variations and interactions. In this study, urinary proteomics of neonatal donkeys were characterized and compared to the profiles of adult donkeys to provide a reference database for healthy neonatal donkeys. T...
Kallikrein proteoforms and reproductive parameters in stallion are conditioned by climate.
Scientific reports    November 4, 2022   Volume 12, Issue 1 18690 doi: 10.1038/s41598-022-21350-w
Senra RL, Ramírez-López CJ, Magalhães-Júnior MJ, Neves JGDS, Barros E, Waddington B, Guimarães SEF, Guimarães JD, Baracat-Pereira MC.Horses are seasonal polyoestrous animals, and the photoperiod is the main factor modulating their reproductive activity. There is no consensus on the andrological and biochemical factors that influence breeding seasonality. To assess the involvement of climate in reproduction, Mangalarga Marchador stallions were monitored over 1 year regarding semen quality and seminal plasma proteome. Here, we show that kallikrein (KLKs) proteoforms in seminal plasma are involved in climate conditioning of reproduction. During the breeding season, greater abundance and different types of KLKs occurred simult...

Pawlas-Opiela M, Jawor P, Galli J, Zak-Bochenek A, Gorczykowski M, Galli J, Sołtysiak Z, Stefaniak T.Infection with Gasterophilus intestinalis (botfly) larvae often occurs in horses. The aim of the study was to isolate the larvae of G. intestinalis and evaluate the serum and salivary humoral immune response using self-developed ELISA in G. intestinalis infected horses. Blood serum or saliva samples were taken from 125 infected horses and 54 uninfected slaughtered horses. The antigens from G. intestinalis larvae were used for development of ELISA in order to evaluate the intensity of G. intestinalis IgG, IgM, and IgA antibody reactivity in the serum or saliva of naturally infected horses and h...
Counts of hyaluronic acid-containing extracellular vesicles decrease in naturally occurring equine osteoarthritis.
Scientific reports    October 20, 2022   Volume 12, Issue 1 17550 doi: 10.1038/s41598-022-21398-8
Mustonen AM, Lehmonen N, Oikari S, Capra J, Raekallio M, Mykkänen A, Paakkonen T, Rilla K, Niemelä T, Nieminen P.Osteoarthritis (OA) is a degenerative joint disease with inadequately understood pathogenesis leading to pain and functional limitations. Extracellular vesicles (EVs) released by synovial joint cells can induce both pro- and anti-OA effects. Hyaluronic acid (HA) lubricates the surfaces of articular cartilage and is one of the bioactive molecules transported by EVs. In humans, altered EV counts and composition can be observed in OA synovial fluid (SF), while EV research is in early stages in the horse-a well-recognized OA model. The aim was to characterize SF EVs and their HA cargo in 19 horses...

Uterine extracellular vesicles as multi-signal messengers during maternal recognition of pregnancy in the mare.
Scientific reports    September 16, 2022   Volume 12, Issue 1 15616 doi: 10.1038/s41598-022-19958-z
Rudolf Vegas A, Hamdi M, Podico G, Bollwein H, Fröhlich T, Canisso IF, Bauersachs S, Almiñana C.In contrast to other domestic mammals, the embryo-derived signal(s) leading to maternal recognition of pregnancy (MRP) are still unknow in the mare. We hypothesize that these embryonic signals could be packed into uterine extracellular vesicles (uEVs), acting as multi-signal messengers between the conceptus and the maternal tract, and contributing to MRP. To unveil these signals, the RNA and protein cargos of uEVs isolated from uterine lavages collected from pregnant mares (P; day 10, 11, 12 and 13 after ovulation) and cyclic control mares (C; day 10 and 13 after ovulation) were analyzed. Our ...
