Topic:Adrenal

The adrenal glands in horses are small endocrine organs located near the kidneys, responsible for producing a variety of hormones essential for physiological regulation. These hormones include cortisol, adrenaline, and aldosterone, which are involved in stress response, metabolism, and electrolyte balance. The functioning of the adrenal glands is integral to the horse's ability to respond to stressors and maintain homeostasis. Research in equine endocrinology often focuses on understanding the regulation of adrenal hormones, their role in equine health, and the impact of disorders such as equine Cushing's disease and adrenal insufficiency. This page compiles peer-reviewed research studies and scholarly articles that explore the anatomy, function, and clinical implications of adrenal gland activity in horses.
Characterization of equine GST A3-3 as a steroid isomerase.
The Journal of steroid biochemistry and molecular biology    November 24, 2017   Volume 178 117-126 doi: 10.1016/j.jsbmb.2017.11.011
Lindström H, Peer SM, Ing NH, Mannervik B.Glutathione transferases (GSTs) comprise a superfamily of enzymes prominently involved in detoxication by making toxic electrophiles more polar and therefore more easily excretable. However some GSTs have developed alternative functions. Thus, a member of the Alpha class GSTs in pig and human tissues is involved in steroid hormone biosynthesis, catalyzing the obligatory double-bond isomerization of Δ-androstene-3,17-dione to Δ-androstene-3,17-dione and of Δ-pregnene-3,20-dione to Δ-pregnene-3,20-dione on the biosynthetic pathways to testosterone and progesterone. The human GST A3-3 is the ...

Differential expression of skeletal muscle genes following administration of clenbuterol to exercised horses.
BMC genomics    August 9, 2016   Volume 17 596 doi: 10.1186/s12864-016-2945-2
Knych HK, Harrison LM, Steinmetz SJ, Chouicha N, Kass PH.Clenbuterol, a beta2-adrenergic receptor agonist, is used therapeutically to treat respiratory conditions in the horse. However, by virtue of its mechanism of action it has been suggested that clenbuterol may also have repartitioning affects in horses and as such the potential to affect performance. Clenbuterol decreases the percent fat and increases fat-free mass following high dose administration in combination with intense exercise in horses. In the current study, microarray analysis and real-time PCR were used to study the temporal effects of low and high dose chronic clenbuterol administr...

Domestic animal endocrinology    June 28, 2016   Volume 56 Suppl S101-S111 doi: 10.1016/j.domaniend.2016.02.008
van der Kolk JH, Fouché N, Gross JJ, Gerber V, Bruckmaier RM.In this review, we address the function of the hypothalamus-pituitary-adrenocortical (HPA) axis with special emphasis on the comparison between the bovine and equine species. The pars intermedia of the pituitary gland is particularly well developed in horses and cattle. However, its function is not well appreciated in cattle yet. The Wulzen's cone of the adenohypophysis is a special feature of ruminants. Total basal cortisol concentration is much higher in horses than that in cows with similar free cortisol fractions. Corticotropin-releasing factor (CRF) concentrations in equine pituitary veno...

Scheidegger MD, Gerber V, Ramseyer A, Schüpbach-Regula G, Bruckmaier RM, van der Kolk JH.The aim of this study was to further characterize the ACTH stimulation test as reflected by salivary cortisol response and to measure the short- and long-term repeatability of it in healthy horses as a tool to assess the capacity of the adrenal cortex to secrete cortisol. Nineteen healthy horses were subjected to 3 ACTH stimulation tests. Intervals were 2 wk and 5 mo between the first and second and the second and third tests, respectively. A dose of 1-μg/kg BW synthetic ACTH was injected intravenously. Saliva samples were collected at baseline and at 30, 60, 90, 120, 150, and 180 min afte...
Fecal Glucocorticoid Analysis: Non-invasive Adrenal Monitoring in Equids.
Journal of visualized experiments : JoVE    April 25, 2016   Issue 110 53479 doi: 10.3791/53479
Yarnell K, Purcell RS, Walker SL.Adrenal activity can be assessed in the equine species by analysis of feces for corticosterone metabolites. During a potentially aversive situation, corticotrophin releasing hormone (CRH) is released from the hypothalamus in the brain. This stimulates the release of adrenocorticotrophic hormone (ACTH) from the pituitary gland, which in turn stimulates release of glucocorticoids from the adrenal gland. In horses the glucocorticoid corticosterone is responsible for several adaptations needed to support equine flight behaviour and subsequent removal from the aversive situation. Corticosterone met...

Other lesions included hepatic fibrosis, erosive gastritis, and bilateral adrenal cortical hyperplasia. Pancréatite interstitielle chronique active comme cause d’une obstruction du côlon transversal et de coliques chez un cheval. Un cheval Quarter horse adulte a été euthanasié après de...
HORSE SPECIES SYMPOSIUM: Glucocorticoid programming of hypothalamic-pituitary-adrenal axis and metabolic function: Animal studies from mouse to horse.
Journal of animal science    October 7, 2015   Volume 93, Issue 7 3245-3260 doi: 10.2527/jas.2014-8612
Jellyman JK, Valenzuela OA, Fowden AL.Adrenal glucocorticoids, such as cortisol, are essential for normal fetal development and for maintaining homeostasis in adults. Developmental studies in humans and other animals have shown that exposure to excess glucocorticoids during critical windows of perinatal development can program permanent changes in hypothalamic-pituitary-adrenal (HPA) axis function and metabolic function, with adverse implications for the long-term health of the exposed offspring. The current review compares the programming of postnatal HPA axis function and glucose homeostasis among different species overexposed p...

Molecular & cellular proteomics : MCP    September 13, 2015   Volume 14, Issue 11 3072-3086 doi: 10.1074/mcp.M115.054635
McGorum BC, Pirie RS, Eaton SL, Keen JA, Cumyn EM, Arnott DM, Chen W, Lamont DJ, Graham LC, Llavero Hurtado M, Pemberton A, Wishart TM.Equine grass sickness (EGS) is an acute, predominantly fatal, multiple system neuropathy of grazing horses with reported incidence rates of ∼2%. An apparently identical disease occurs in multiple species, including but not limited to cats, dogs, and rabbits. Although the precise etiology remains unclear, ultrastructural findings have suggested that the primary lesion lies in the glycoprotein biosynthetic pathway of specific neuronal populations. The goal of this study was therefore to identify the molecular processes underpinning neurodegeneration in EGS. Here, we use a bottom-up approach be...

Pre-analytical stability of adrenocorticotrophic hormone from healthy horses in whole blood, plasma and frozen plasma samples.
Veterinary journal (London, England : 1997)    February 11, 2015   Volume 204, Issue 1 123-124 doi: 10.1016/j.tvjl.2015.02.010
Prutton JS, Kass PH, Watson JL, Pusterla N.The stability of equine adrenocorticotrophic hormone (ACTH) in blood samples is not fully known. The study objectives were to determine ACTH stability (1) in whole blood and plasma over 72 h at either 4 or 21 °C, and (2) in plasma frozen at either -20 or -80 °C over 30 days. Nine horses were sampled and ACTH concentration were measured after storage as whole blood or plasma, at 4, 21, -20 and -80 °C for up to 30 days. The ACTH concentration was significantly reduced at 24 h but remained stable when plasma was frozen at -20 and -80 °C for 30 days. Beyond 24 h, samples stored a...
Concomitant inhibition of primary equine bronchial fibroblast proliferation and differentiation by selective β2-adrenoceptor agonists and dexamethasone.
European journal of pharmacology    August 14, 2014   Volume 741 205-213 doi: 10.1016/j.ejphar.2014.07.056
Franke J, Abraham G.Altered airway cell proliferation plays an important role in the pathogenesis of human bronchial asthma and chronic obstructive pulmonary disease (COPD) as well as the equine recurrent airway obstruction (RAO) with consistent changes, i.e. narrowing the airway wall, explained by proliferation and differentiation of fibroblasts. In permanent cell lines, it has been suggested that β2-adrenoceptor agonists and glucocorticoids regulate cell proliferation via the β2-adrenoceptor pathway; indeed, no study was carried out in fresh isolated primary equine bronchial fibroblasts (EBF). We characterize...
Neonatal glucocorticoid overexposure programs pituitary-adrenal function in ponies.
Domestic animal endocrinology    July 3, 2014   Volume 50 45-49 doi: 10.1016/j.domaniend.2014.06.006
Jellyman JK, Valenzuela OA, Allen VL, Forhead AJ, Holdstock NB, Fowden AL.The present study tested the hypothesis that overexposure to endogenous glucocorticoids in neonatal life alters the reactivity of the hypothalamic-pituitary-adrenal (HPA) axis in ponies at 1 and 2 yr of age. Newborn foals received saline (0.9% NaCl, n = 8, control) or long-acting adrenocorticotropic hormone (ACTH1-24) (Depot Synacthen 0.125 mg intramuscularly twice daily, n = 9) for 5 d after birth to raise cortisol concentrations 5- to 6-fold. At 1 and 2 yr of age, HPA axis function was assessed by bolus administration of short-acting ACTH1-24 (1 μg/kg intravenous) and insulin (0.5 U/kg...

Parturition in horses is dominated by parasympathetic activity of the autonomous nervous system.
Theriogenology    March 27, 2014   Volume 82, Issue 1 160-168 doi: 10.1016/j.theriogenology.2014.03.015
Nagel C, Erber R, Ille N, von Lewinski M, Aurich J, Möstl E, Aurich C.External and internal stressors prolong parturition in different species. At parturition, sympathoadrenal activation should be avoided because an increased sympathetic tone may cause uterine atonia via β2-receptors. We hypothesized that at physiological parturition, horses are under parasympathetic dominance, and stress-response mechanisms are not activated during delivery of the foal. To evaluate stress responses, heart rate, heart rate variability, catecholamines, and cortisol were analyzed in mares (n = 17) throughout foaling. Heart rate decreased from 2 hours before (51 ± 1 beats/minute)...

European journal of pharmacology    October 10, 2013   Volume 720, Issue 1-3 363-375 doi: 10.1016/j.ejphar.2013.10.003
Hikasa Y, Masuda K, Asakura Y, Yamashita Y, Sato C, Kamio M, Miura A, Taniguchi T, Minamizuru N.This study aimed to pharmacologically identify and characterize α2-adrenoceptors and imidazoline (I) receptors (I1- and I2-subtype) on canine, feline, bovine, equine, murine, and leporine platelet membranes. Saturation binding studies with both (3)H-yohimbine and (3)H-clonidine showed that α2-adrenoceptors were expressed on canine, leporine, feline, and murine platelets but not on bovine and equine platelets. In competition studies, the rank order of affinity of 6 compounds for canine platelet α2-adrenoceptors was similar to that of potency at α2A-subtype reported in human platelets. Satur...
Response of the hypothalamic-pituitary-adrenal axis to stimulation tests before and after exercise training in old and young Standardbred mares.
Journal of animal science    September 17, 2013   Volume 91, Issue 11 5208-5219 doi: 10.2527/jas.2013-6329
Liburt NR, McKeever KH, Malinowski K, Smarsh DN, Geor RJ.This study tested the hypotheses that age-induced alteration in cortisol, ACTH, and glucose concentrations are due to differences in the response of the hypothalamic-pituitary-adrenal axis and that exercise training would attenuate these differences. Six old (22.0±0.7 yr; mean±SE) and 6 young (7.3±0.6 yr) unfit Standardbred mares ran 3 graded exercise tests (GXT): before (GXT1), after 8 wk of training (GXT2), and at study end at 15 wk (GXT3). Mares trained 3 d/wk at 60% maximum heart rate. Each mare underwent 5 endocrine stimulation tests pre- and posttraining: 1) control (CON), 2) adrenoco...
Expression of β2 adrenoceptors within enteric neurons of the horse ileum.
Research in veterinary science    August 12, 2013   Volume 95, Issue 3 837-845 doi: 10.1016/j.rvsc.2013.07.006
Bombardi C, Grandis A, Gardini A, Sorteni C, Clavenzani P, Chiocchetti R.The activity of the gastrointestinal tract is regulated through the activation of adrenergic receptors (ARs). Since data concerning the distribution of ARs in the horse intestine is virtually absent, we investigated the distribution of β2-AR in the horse ileum using double-immunofluorescence. The β2-AR-immunoreactivity (IR) was observed in most (95%) neurons located in submucosal plexus (SMP) and in few (8%) neurons of the myenteric plexus (MP). Tyrosine hydroxylase (TH)-IR fibers were observed close to neurons expressing β2-AR-IR. Since β2-AR is virtually expressed in most neurons located...
