Aggregation-associated loss of antigenicity observed for denatured virion protein 1 of Equine rhinitis A virus in an enzyme-linked immunosorbent assay.
Virus research    March 20, 2009   Volume 143, Issue 1 130-133 doi: 10.1016/j.virusres.2009.03.003
Kriegshäuser G, Kuechler E, Skern T.Equine rhinitis A virus (ERAV) is a picornavirus which causes an acute respiratory infection in horses worldwide, and virus neutralization (VN) has been the standard method for the detection of ERAV antibody in horse serum. Previous studies have identified recombinant virion protein VP1 (rVP1) purified under native conditions to be of high potential for the development of a diagnostic ERAV enzyme-linked immunosorbent assay (ELISA). This study presents an optimized protocol for the expression and purification of native full-length rVP1. Furthermore, we demonstrate that, upon denaturation, rVP1 ...
Refinement of a quantitative trait locus on equine chromosome 5 responsible for fetlock osteochondrosis in Hanoverian warmblood horses.
Animal genetics    March 20, 2009   Volume 40, Issue 4 553-555 doi: 10.1111/j.1365-2052.2009.01865.x
Lampe V, Dierks C, Distl O.In this report, we provide 29 new informative microsatellites distributed over a region of 21 Mb on horse chromosome (ECA) 5 and refine a quantitative trait locus (QTL) for fetlock osteochondrosis dissecans (OCD) to a genome-wide significant interval between 78.03 and 90.23 Mb on ECA5. Genotyping was performed in 211 Hanoverian warmblood horses from 14 paternal half-sib groups. Within this OCD-QTL, collagen type XXIV alpha 1 was identified as a potential functional candidate gene for equine osteochondrosis. This report is a further step towards unravelling the genes that cause equine osteochon...
Regulation of platelet activating factor-induced equine platelet activation by intracellular kinases.
Journal of veterinary pharmacology and therapeutics    March 18, 2009   Volume 32, Issue 2 189-196 doi: 10.1111/j.1365-2885.2008.01020.x
Brooks AC, Menzies-Gow NJ, Wheeler-Jones CP, Bailey SR, Elliott J, Cunningham FM.Lipopolysaccharide (LPS) can activate equine platelets directly or indirectly, via leukocyte-derived platelet activating factor (PAF). Thromboxane (Tx) production by LPS-stimulated equine platelets requires p38 MAPK and this kinase has been suggested as a therapeutic target in endotoxaemia. The present study has utilised selective inhibitors to investigate the role of p38 MAPK and two other kinases, phosphatidylinositol-3 kinase (PI3K) and protein kinase C (PKC), in regulating PAF-induced Tx production, aggregation and 5-HT release in equine platelets, and the modification of these responses b...
Adult-young ratio, a major factor regulating social behaviour of young: a horse study.
PloS one    March 18, 2009   Volume 4, Issue 3 e4888 doi: 10.1371/journal.pone.0004888
Bourjade M, de Boyer des Roches A, Hausberger M.Adults play an important role in regulating the social behaviour of young individuals. However, a few pioneer studies suggest that, more than the mere presence of adults, their proportions in social groups affect the social development of young. Here, we hypothesized that aggression rates and social cohesion were correlated to adult-young ratios. Our biological model was naturally-formed groups of Przewalski horses, Equus f. przewalskii, varying in composition. Results: We investigated the social interactions and spatial relationships of 12 one- and two-year-old Przewalski horses belonging to ...
Isolation of Campylobacter fetus subspecies fetus in a two-year-old quarterhorse with chronic diarrhea of an undetermined etiology. Hurcombe SD, Fox JG, Kohn CW.A 2-year-old quarterhorse was evaluated for chronic diarrhea and weight loss of 5 weeks duration after numerous diagnostic tests failed to identify an underlying cause. Historically, the horse was housed at pasture where human household waste vehicles were routinely cleaned and the effluent could run onto the field. Physical examination revealed poor body condition and frequent high-volume diarrhea. Diagnostic testing for Salmonella spp., endoparasites, Cryptosporidium spp., Clostridium spp., and diffuse infiltrative bowel disease were negative. Rectal tissue histopathology failed to identify ...
Field evaluation of a multiplex real-time reverse transcription polymerase chain reaction assay for detection of Vesicular stomatitis virus. Wilson WC, Letchworth GJ, Jiménez C, Herrero MV, Navarro R, Paz P, Cornish TE, Smoliga G, Pauszek SJ, Dornak C, George M, Rodriguez LL.Sporadic outbreaks of vesicular stomatitis (VS) in the United States result in significant economic losses for the U.S. livestock industries because VS is a reportable disease that clinically mimics foot-and-mouth disease. Rapid and accurate differentiation of these 2 diseases is critical because their consequences and control strategies differ radically. The objective of the current study was to field validate a 1-tube multiplexed real-time reverse transcription polymerase chain reaction (real-time RT-PCR) assay for the rapid detection of Vesicular stomatitis New Jersey virus and Vesicular st...

Revue scientifique et technique (International Office of Epizootics)    March 17, 2009   Volume 27, Issue 3 893-897 doi: 10.20506/rst.27.3.1846
Cano A, Galosi CM, Martin Ocampos GP, Ramirez GC, Vera VJ, Villamil LC, Chaparro JG.This paper describes the isolation and characterisation of equine herpesvirus 1 (EHV-1) in Colombia. The virus was isolated from a nasal swab and an aborted foetus of a pregnant mare imported from Argentina, with clinical signs of rhinopneumonitis. The new strain was characterised through culture and morphological, serological and immunocytochemical studies. Polymerase chain reaction and DNA restriction maps revealed an EHV-1 1P genome. This is the first report on the isolation and characterisation of EHV-1 in Colombia.
Young foal and adult horse monocyte-derived dendritic cells differ by their degree of phenotypic maturity.
Veterinary immunology and immunopathology    March 14, 2009   Volume 131, Issue 1-2 1-8 doi: 10.1016/j.vetimm.2009.03.002
Mérant C, Breathnach CC, Kohler K, Rashid C, Van Meter P, Horohov DW.Newborn foals are very susceptible to infections by opportunistic pathogens such as Rhodococcus equi. This susceptibility is thought to be due to the immaturity of their immune system, in particular their inability to produce interferon-gamma. This deficiency may result from an insufficiency in accessory signals. We therefore compared monocyte-derived dendritic cells (MoDC) from foals and from adult horses. CD172, MHC-I and MHC-II were generally expressed on more than 90% MoDC from foals and adults. CD1w2(+)CD86(+) cells tended to be less represented in 2-3-week-old foals than in adults. This ...

Arthritis research & therapy    March 9, 2009   Volume 11, Issue 2 R35 doi: 10.1186/ar2640
de Grauw JC, van de Lest CH, van Weeren PR.Inflammation is an important feature of many joint diseases, and levels of cartilage biomarkers measured in synovial fluid may be influenced by local inflammatory status. Little is known about the magnitude and time course of inflammation-induced changes in cartilage tissue turnover as measured in vivo by synovial fluid markers. We aimed to study temporal changes in concentrations of inflammatory mediators, matrix metalloproteinase activity and cartilage biomarkers over 1 week in joints with experimentally induced inflammation. Methods: Localized inflammation was induced in the intercarpal joi...
Direct injection liquid chromatography/electrospray ionization mass spectrometric horse urine analysis for the quantification and confirmation of threshold substances for doping control. II. Determination of theobromine.
Rapid communications in mass spectrometry : RCM    March 6, 2009   Volume 23, Issue 7 1020-1028 doi: 10.1002/rcm.3967
Vonaparti A, Lyris E, Panderi I, Koupparis M, Georgakopoulos C.In equine sport, theobromine is prohibited with a threshold level of 2 microg mL(-1) in urine, hence doping control laboratories have to establish quantitative and qualitative methods for its determination. Two simple liquid chromatography/mass spectrometry (LC/MS) methods for the identification and quantification of theobromine were developed and validated using the same sample preparation procedure but different mass spectrometric systems: ion trap mass spectrometry (ITMS) and time-of-flight mass spectrometry (TOFMS). Particle-free diluted urine samples were directly injected into the LC/MS ...
Identification of Bartonella henselae in an aborted equine fetus.
Veterinary pathology    March 6, 2009   Volume 46, Issue 2 277-281 doi: 10.1354/vp.46-2-277
Johnson R, Ramos-Vara J, Vemulapalli R.This report describes the characterization of a Bartonella henselae abortion in an equine fetus by gross, histologic, immunohistochemical, ultrastructural, and molecular methods. Bartonella henselae can cause cat scratch disease, bacillary angiomatosis, bacillary peliosis, and endocarditis in humans and other animals. The bacterium has been isolated from several mammalian species but only recently from equids; however, it has not been linked to abortion in equids. An aborted equine fetus exhibited necrosis and vasculitis in multiple tissues, with intralesional Gram-negative short-to-spirillar ...

Veterinary ophthalmology    March 6, 2009   Volume 12, Issue 2 125-131 doi: 10.1111/j.1463-5224.2008.00688.x
McMullen RJ, Clode AB, Gilger BC.Ocular photographs are an increasingly important method of documenting lesions for inclusion in medical records, teaching purposes, and research. Availability of affordable high-quality digital imaging equipment has allowed for enhanced capture of desired images because of immediate on-camera viewing and editing. Conversion of the standard digital camera sensor to one that is sensitive to infrared light (i.e. > 760 nm < 1 mm wavelength) can be done inexpensively. In the equine eye, advantages of infrared digital photography include increased contrast of anterior segment images, identific...

Australian veterinary journal    February 28, 2009   Volume 87, Issue 3 110-112 doi: 10.1111/j.1751-0813.2009.00392.x
Harland S, Smith C, Mogg T, Horadagoda N, Dart A.A mare was referred for further evaluation of a mass found in the left caudal abdomen during a routine postpartum reproductive palpation. The mare was clinically normal with no history of health problems. Ultrasonographic examination of the mass confirmed its presence, but the origin of the mass could not be accurately determined. Routine haematology and serum biochemistry results were within normal limits. The mare was initially treated conservatively with antibiotics, but the mass continued to increase in size, so it was surgically excised. The mass involved the left ovary. The mare showed t...
Magnetic resonance imaging of the initial active stage of equine laminitis at 4.7 T. Arble JB, Mattoon JS, Drost WT, Weisbrode SE, Wassenaar PA, Pan X, Hunt RJ, Belknap JK.Equine laminitis is a severely debilitating disease. There is a poor understanding of the underlying pathophysiology, and traditional imaging modalities have limited diagnostic capacity. High field strength magnetic resonance (MR) imaging allows direct visualization of the laminae, which other modalities do not. This would prove useful both in assessment of clinical patients and in further investigation into the pathophysiology of the disease. The objective of this study was to characterize the anatomic changes within the equine foot associated with the initial active stage of laminitis. Image...

Twenty horses were studied and their medical records and MR images were reviewed retrospectively. Long-term follow-up information was obtained by telephonic questionnaires of owners, trainers, or referring veterinarians. Desmopathy of the medial collateral ligament (80%) and enthesopathy of the affected collateral ligament (80%) were common MR imaging features. Treatment consisted of stall rest followed...
Ultrasonographic assessment of laryngohyoid position as a predictor of dorsal displacement of the soft palate in horses. Chalmers HJ, Yeager AE, Ducharme N.Dorsal displacement of the soft palate is an important cause of poor performance in racehorses, yet its etiology is not fully understood. Diagnosis requires treadmill videoendoscopy, which is not widely available. The relationship of the larynx, the hyoid apparatus, and the remainder of the skull may be important in predisposing horses to dorsal displacement of the soft palate. We hypothesized that this relationship could be accurately assessed in unsedated horses through ultrasonographic examination. Fifty-six racehorses presented for evaluation of poor performance were subjected to treadmill...
